3D culture systems as a tool for studying dendritic cells

Abstract:

Dendritické buňky řadíme mezi buňky bílé krevní řady a považujeme je za přechodový článek mezi specifickou a nespecifickou imunitou. Jsou jako jedny z nejspolehlivějších schopny prezentace antigenu dalším buňkám směrem z periferie do lymfatických uzlin. V současné době podléhají intenzivnímu výzkumu, a to hned v několika oblastech vědy s přesahem do terapie infekčních onemocnění (včetně nemoci HIV), terapie autoimunitních onemocnění, problematiky transplantací, či podpoře protinádorové imunity. V rámci stále se vyvíjejícího výzkumu alternativních metod k pokusům na zvířatech se rozvíjejí také experimenty s tkáňovými kulturami, které by mohly pomoci snížit počty potřebných kusů živých zvířat ve výzkumu. I tyto metody se stále vyvíjejí a je nutné je zdokonalovat pro co možná největší autenticitu a napodobení prostředí organismu. Práce se zabývá 3D systémy kultivace dendritických buněk pomocí nanovláknových scaffoldů pocházejících z biodegradovatelného plastu polykaprolaktonu a také fibroinu, který produkují některé čeledi hmyzu, včetně bource morušového (Cricula trifenestrata). Po provedení experimentů z praktické části bylo zjištěno, že nejnižší výskyt buněk po kultivaci byl nalezen na prostých polykaprolaktonových vláknech. Naopak narůstající počet buněk byl obsažen na vláknech se zvyšujícím se procentuálním obsahem fibroinu.

Abstract:

Dendritic cells are classified as white blood cells and we consider them to be a transitional link between specific and non-specific immunity. They are one of the most reliable capable of antigen presentation to other cells from the periphery to the lymph nodes. Currently, they are subject to intensive research in several areas of science with an overlap into the therapy of infectious diseases (including HIV), the therapy of autoimmune diseases, the issue of transplantation, or the support of anti-tumor immunity. As part of the ever-evolving research into alternative methods to animal testing, tissue culture experiments are also being developed that could help reduce the number of live animals needed in research. Even these methods are still developing and it is necessary to improve them for the greatest possible authenticity and imitation of the organims´s environment. The work deals with 3D systems of dendritic cell cultivation using nanofibrous scaffolds originating from the biodegradable plastic polycaprolactone as well as fibroin, which are produced by some families of insects, including the silkworm (Cricula trifenestrata). After experiments from the practical part, it was found that the lowest occurrence of cells after cultivation was found with plain polycaprolactone fibers. On the contrary, the increasing number of cells was contained on fibers with a percentage of fibroin content.
