Isolation of Biogenic Amine-Degrading Microorganisms

Abstract:

Bakalářská práce je zaměřena na izolaci mikroorganizmů degradujících biogenní aminy. Teoretická část se zabývá biogenními aminy, jejich vznikem, významem v potravinách a mikroorganizmy schopných jejich degradace. V praktické části této práce bylo izolováno a identifikováno pomocí metody MALDI-TOF MS 17 degradérů biogenních aminů. U všech izolátů byla také ověřována schopnost redukce vybraných biogenních aminů po 24 a 48 hodinové kultivaci v minerálním médiu s biogenními aminy a v minerálním médiu s biogenními aminy a glukózou pomocí vysokoúčinné kapalinové chromatografie s UV detekcí po předchozí derivatizaci dansylchloridem. Z výsledků je patrné, že nejlepším degradérem byl Micrococcus lutes KS15. Došlo k poklesu koncentrace všech sledovaných biogenních aminů nejméně o 70 % a nejlépe pak degradoval histamin, jehož konečná hodnota činila 5 % po 24 hodinách kultivace v minerálním médiu s biogenními aminy.

Abstract:

The thesis focuses on the isolation of biogenic amine-degrading microorganisms. The theoretical part is focused on biogenic amines, their origin, importance in food and microorganisms able to degrade them. In the practical part of this work were isolated 17 microorganisms with degradation activity of biogenic amines. These microorganisms were identified by MALDI-TOF mass spectrometry. In the following experiment it was monitored degradation of selected biogenic amines after 24 and 48 hour growth in the mineral medium with biogenic amines and in the mineral medium with biogenic amines and glucose. The loss of biogenic amines was analyzed by the high performance liquid chromatography with UV detection after derivatization with dansylchloride. The results show that the best aminedegrading microorganism was Micrococcus luteus KS15. All of the biogenic amines were degraded by 70%. The lowest concentration was detected at histamine. The final value was 5% af-ter 24 hour growth in the mineral medium with biogenic amines.
