Addictive substances in hair

Abstract:

Návykové látky jsou věčným problémem naší společnosti. Působí nepříznivě na náš organismus a mohou mít letální následky. Pro průkaz návykových látek ve vlasech je potřeba najít ve vzorku i jejich metabolit. Metabolit delta-9-hydrocanabinol je kyselina 11-nor-delta-9-tetrahydrocannabinolová, u metamfetaminu se jedná o amfetamin, u metylendioxymetamfetaminu to je metylendioxyamfetamin. Tato bakalářská práce se zaměřuje na záchyt a stanovení těchto látek ve vlasech a zvolení nejvhodnější metody pro každý analyt. Analýza byla provedena na vlasech od živých i zemřelých osob. Vlasová analýza umožňuje rozšířit detekční okno, v závislosti na délce vlasů, a poskytnout informace o dlouhodobém užívání zmíněných látek jako důkazní materiál např. u soudu. Pro amfetaminy byla za nejvhodnější metodu uvolnění analytů z vlasů zvolena hydrolýza pufrem a následná analýza kapalinovou chromatografií s hmotnostním spektrometrem. Hodnota limitu detekce amfetaminů byla stanovena na 7,5 pg/mg a limit kvantifikace na 25 pg/mg. Jako nejvhodnější metoda pro vyvázání nox z vlasů u kanabinoidů se ukázala alkalická hydrolýza s následnou analýzou plynovou chromatografií s hmotnostním spektrometrem. Hodnota limitu detekce u delta-9-hydrocanabinol byla stanovena na 2,4 pg/mg a hodnota limitu kvantifikace 8 pg/mg, u kyseliny-11-nor-delta-tetrahydrocannabinolové byla hodnota limitu detekce stanovena na 0,51 pg/mg a hodnota limitu kvantifikace na 1,7 pg/mg. Byly provedeny kalibrační křivky a opakovatelnost metod.

Abstract:

Addictive substances are a everlasting problem our society. They cause unfavorably on our organism and can have lethal consequences. In order to detect addictive substances in the hair, it is also necessary to find their metabolite in the sample. Metabolite delta-9-hydrocannabinol is nor-11-delta-9-tetrahydrocannabinolic acid, for metamphetamin it is amphetamin, for metylendioxymetamphetamin it is metylendioxyamphetamin. This bachelor's thesis focuses on the capture and determination of these substances in hair and the selection of the most suitable method for each analyte. The analysis was carried out on hair from living and deceased persons. Hair analysis makes it possible to expand the detection window, depending on the length of the hair, and to provide information about the long-term use of the mentioned substances as evidence, e.g. in court. For amphetamines, hydrolysis with a buffer and subsequent analysis by liquid chromatography with a mass spectometer was chosen as the most suitable method of analyte release from hair. The value of the limit of detection of amphetamines was set at 7.5 pg/mg and the limit of quantification at 25 pg/mg. Alkaline hydrolysis proved to be the most suitable method for untie tied up nox from hair by cannabinoids with subsequent analysis by gas chromatography with a mass spectrometer. The value of the limit of detection of delta-9-hydrocannabinol was set at 2.4 pg/mg and the limit of quantification was 8 pg/mg, for 11-nor-delta-9-tetrahydrocannabinol acid the limit of detection was set at 0.51 pg/mg and limit of quantification at 1.7 pg/mg. Calibration curves and repeatability of the metods were performed.
