Assessment of microbial contamination in different species of honey

Abstract:

Diplomová práce se zaměřuje na mikrobiální kontaminaci různých druhů medů dostupných na trhu. Cílem práce bylo prozkoumat rozsah mikrobiální kontaminace medu (celkem 5 vzorků) a identifikovat mikroorganismy přítomné v těchto vzorcích pomocí různých metod (kultivace na různých půdách, kataláza a oxidáza test, PCR, RAPD, sekvenování DNA). Ze získaných výsledků bylo zjištěno, že z 51 vzorků izolátů se jednalo o kataláza pozitivní (34) a negativní (17); oxidáza pozitivní (30) a negativní (21); gram-negativní (10) a gram-pozitivní (36); bakterie (46) a kvasinky (5). Analýze sekvenování bylo podrobeno 13 vzorků izolátů, kdy nejčastěji detekovanými mikroorganismy byly Bacillus spp. (69,2 %), Rhodotorula spp. (23,1 %) a Pseudomonnas spp. (7,7 %). Zjištěné mikroorganismy byly potvrzené jako běžně se vyskytují mikroorganismy v trávicím traktu včel či jejich prostředí.

Abstract:

The thesis focuses on microbial contamination of various types of honey available on the market. The aim of the thesis was to investigate the extent of microbial contamination of honey (5 samples in total) and to identify microorganisms present in these samples using different methods (cultivation on different soils, catalase and oxidase test, PCR, RAPD, DNA sequencing). From the results obtained, it was found that out of 51 isolate samples, they were catalase positive (34) and negative (17); oxidase positive (30) and negative (21); gram-negative (10) and gram-positive (36); bacteria (46) and yeasts (5). Sequencing analysis was carried out on 13 isolate samples, with Bacillus spp. (69.2 %), Rhodotorula spp. (23.1 %) and Pseudomonnas spp. (7.7 %). The microorganisms detected were confirmed as commonly occurring microorganisms in the digestive tract of bees or their environment.
