Methylation of plant telomeres

Abstract:

Mezi epigenetické modifikace řadíme modifikace bazí nukleových kyselin a modifikace histonů. K nejvíce prostudované epigenetické modifikaci patří metylace DNA, v eukaryotických buňkách se vyskytuje převážně metylace na pátém uhlíku cytosinu. Mezi metody studující metylaci DNA řadíme vysokoúčinné separace jako je HPLC a HPCE. Využívány jsou také restrikční endonukleázy, citlivé na metylovaný cytosin ve svých restrikčních místech. Řada přístupů je založena na konverzi DNA bisulfitem a následné amplifikaci studovaného úseku pomocí PCR. K získání kompletního metylačního obrazu DNA daného organismu (metylomu) jsou používány metody celogenomového sekvenování. Metylaci cytosinů lokalizovaných v rostlinných telomerových sekvencích nelze analyzovat konvenčními přístupy, kromě celogenomových technik. V práci (Vrbský 2010) byla navržena metoda analýzy metylace telomerových cytosinů, kdy je bisulfitem konvertovaná genomová DNA přenesena na nylonovou membránu a následně hybridizuje se specifickými radioaktivně značenými oligonukleotidovými sondami. Touto metodou jsme studovali metylaci cytosinů lokalizovaných v telomerických repetitivních motivech AAATCCC v rostlinách Arabidopsis thaliana divokého typu a mutantních liniích nesoucích inzerci T-DNA v genech klíčových pro udržení stabilního metylačního obrazu. Zjistili jsme, že množství metylovaných cytosinů v telomerách mutantních linií je ve srovnání s rostlinami divokého typu významně nižší. Vzhledem k přítomnosti telomerových repetic i v polohách mimo konce chromozómů v genomu A. thaliana budou výsledky potvrzeny nezávislou metodou.

Abstract:

Epigenetic modifications involve two main mechanisms, modifications of nucleic acids and histone proteins. DNA methylation is the best known epigenetic modification. Variety of techniques is utilized to determine level of methylated cytosines including high-performance separations (HPLC and HPCE), and methylation sensitive restriction endonucleases. Frequently used approaches are based on conversion of non-methylated cytosines by bisulfite followed by amplification of the analyzed region. To determine the whole genome methylation high-throughput sequencing methods are used. Methylation of cytosines located in the plant telomeres is not possible to be analyzed by conventional approaches, except of genome-wide techniques. In the work (Vrbský 2010) sophisticated method for analysis of telomere cytosines methylation was proposed. According to this protocol, bisulfite-converted genomic DNA was transferred to nylon membrane and hybridized against specific radioactively labeled oligonucleotide probes. We used this method for analyzing methylation of telomeric cytosines in wild type and mutant lines of Arabidopsis thaliana plant. Mutant lines contain T-DNA insertion in genes which play key role in the maintenance of stable methylation pattern. We found that the level of 5-methylcytosines in telomeres of mutant lines is significantly lower as compared to wild types plants. Because telomeric repetitive sequences are located even outside of chromosome ends in the A. thaliana genome, results will be confirmed by the independent method.
