Molecular role of catechol-O-methyltransferase in triple negative breast cancer

Abstract:

The bachelor thesis focuses on the effect of catechol-O-methyltransferase (COMT) on the molecular mechanisms of tumorigenesis in triple negative breast cancer (TNBC). COMT has been proposed as a potential therapeutic target in TNBC based on its pro-migratory role and promotion of invasive cell phenotype in previous studies. Theoretical part of the thesis is devoted to the characterization of COMT enzyme, its tumour-suppressor role on estrogen-positive breast tumours, and recent findings in the literature in which COMT enzyme appears to have an opposing role, i.e., a pro-tumorigenic role. At the end of the theoretical part the CRISPR-Cas9 method is described, which served as a platform for knock-down of COMT expression in TNBC cells. Experimental part describes the proteome changes and enriched pathways associated with COMT knock-down in MDA-MB-231 cell line represented by three clones, D10, F11 and B1. Proteomics data showed the impact of COMT knock-down on the levels of proteins related to tumorigenesis and invasiveness in TNBC tumor cells, i.e. proteins regulating migration, motility, adhesiveness, angiogenesis and proliferation. Thus, we confirmed by proteomics analysis of the TNBC model that COMT supports tumorigenesis and has a potentially oncogenic role in TNBC.

Abstract:

Bakalářská práce se zabývá studiem vlivu enzymu katechol-O-methyltransferasy (COMT) na molekulární mechanismy tumorigeneze u trojitě-negativního subtypu karcinomu prsu (TNBC). COMT byl v předchozích studiích navržen jako potenciální terapeutický cíl u TNBC na základě jeho pro-migrační úlohy a podpory invazivního fenotypu buněk. Teoretická část práce se věnuje charakteristice enzymu COMT, jeho tumor supresorovým účinkům u estrogen-pozitivních karcinomů prsu a aktuálním poznatkům v literatuře, v rámci kterých enzym COMT jeví opačnou roli, tj. protumorigenní. Na konci teoretické části je představena metoda CRISPR/Cas9, která byla použita pro umlčení exprese COMT v buňkách TNBC. Experimentální část se věnuje změnám proteomu a obohaceným drahám spojeným s umlčením exprese COMT v buňkách linie MDA-MB-231 zastoupených třemi klony D10, F11 a B1. Proteomická data ukazují vliv umlčení COMT na hladiny proteinů souvisejících s podporou tumorigeneze nádorových buněk TNBC, tj. proteinů regulujících migraci, motilitu, adhezivitu, angiogenezi a proliferaci buněk. Proteomickou analýzou TNBC modelu jsme tedy potvrdili, že enzym COMT stojí za podporou tumorigeneze a má potenciálně onkogenní úlohu u TNBC.
