Abstract:
Diploma thesis is focused on preparation and characterization of the product of gene 3133 from bacterium Paracoccus denitrificans. The gene 3133 codes an enzyme NADPH:FMN oxidoreductase. Kinetical parameters of the enzyme, mechanism of enzymatic reaction, dependency of enzyme activity on reaction conditions and covalent modification of amino acid groups by chemical reagents were measured. Diplomová práce se zabývá přípravou a charakterizací produktu genu 3133 z baktérie Paracoccus denitrificans. Gen 3133 kóduje enzym NADPH:FMN oxidoreduktasu. U enzymu byly stanoveny kinetické parametry, mechanizmus reakce, závislost aktivity na podmínkách reakce a kovalentní modifikací aminokyselinových skupin enzymu chemickými činidly.
