Abstract:

Diplomová práce se zabývá vlivem symbiotických interakcí mezi rostlinou a půdními mikroorganismy při stresu suchem. Rostlina čelí celé řadě biotických a abiotických faktorů. Sucho patří k nejzávažnějšímu stresu, který se na rostlinách negativně projevuje zejména ve snížení růstu, výnosu a kvalitě plodin. Jedním z adaptačních mechanismů rostliny při stresu suchem je navázání vzájemně prospěšného symbiotického vztahu s půdními mikroorganismy. Mezi hlavní symbiotické mikroorganismy se řadí skupina bakterie podporující růst rostlin (PGPR) a arbuskulární mykorhizní houby. Pro navázání symbiotického vztahu rostlina vylučuje do půdy chemoatraktivní látky, které působí na výše zmíněné skupiny mikroorganismů a přitahují si je do své blízkosti. V praktické části této práce byla ověřená hypotéza, že při stresu suchem dochází k většímu přiblížení mikroorganismů do okolí kořenů než při dobrých zavlažovacích podmínkách. Cílem bylo optimalizovat metodiku sledování symbiotických interakcí ve sterilní a nesterilní (symbiotické) půdě při stresu suchem a zjistit vliv sterilní půdy na růst rostliny. Pro sledování půdních mikroorganismů byly použity kultivační a molekulární metody (qPCR). Fyziologický stav rostliny při stresu suchem byl hodnocen na morfologické a molekulární úrovni.

Abstract:

The diploma thesis deals with the effect of symbiotic interactions between plant and soil microorganisms during drought stress. The plant faces a range of biotic and abiotic factors. Drought is one of the most severe stresses that negatively affects plants, especially in terms of reduced growth, yield and crop quality. One of the plant's adaptation mechanisms to drought stress is to establishing a mutually beneficial symbiotic relationship with soil microorganisms. The main symbiotic microorganisms include the plant growth promoting bacteria (PGPR) group and arbuscular mycorrhizal fungi. In order to establish a symbiotic relationship, the plant secretes chemoattractants, into the soil, that act on the above groups of microorganisms and attract them to their vicinity. In the practical part of this thesis, the hypothesis that under drought stress, there is a greater approach of microorganisms to the vicinity of the roots than under good irrigation conditions was tested. The aim was to optimising the methodology for monitoring symbiotic interactions in sterile and non-sterile (symbiotic) soil under drought stress and to determine the effect of sterile soil on plant growth. Cultivation and molecular methods (qPCR) were used to monitor soil microorganisms. The physiological state of the plant under drought stress was evaluated at morphological and molecular levels.
