Cloning and characterisation of glycosyltransferases from human pathogen Aspergillus fumigatus

Abstract:

Předložená práce je zaměřena na studium glykosyltransferas, konkrétně na β-1,4-mannosyltransferasu z lidského patogenu A. fumigatus. GTs se významně podílejí na stavbě buněčné stěny patogenních organismů a tím i na jejich virulenci. Proto je získání informací o těchto enzymech velmi důležité. Příprava GTs je velmi obtížná, protože se téměř vždy jedná o transmembránové proteiny, je tedy nutné optimalizovat podmínky pro jejich přípravu. Práce je zaměřena na β-1,4-mannosyltransferasu (Alg1) a její katalytickou doménu. Oba geny byly vklonovány do vektoru pET29b. Celý protein Alg1 se nepodařilo připravit, přestože byly vyzkoušeny různé podmínky optimalizace. Katalytická doména proteinu Alg1 byla po optimalizaci připravena v buněčném kmenu E. coli BL21 (DE3). Téměř všechen cílový protein byl produkován do nerozpustných inkluzních tělísek, proto byla snaha purifikovat katalytickou doménu proteinu Alg1 za denaturačních podmínek. Vyzkoušeny byly také nativní podmínky pro purifikaci, protože v rozpustné složce se nacházelo také malé množství proteinu. Byly vyzkoušeny tři různé sorbenty pro purifikaci za využití chelatační chromatografie – Ni-NTA, Cu-NTA a Cu-IDA. Cílový protein se nepodařilo purifikovat na žádné z nich, i přesto, že hmotnostní analýzou byla potvrzena přítomnost oligohistidinové kotvy. Budoucnost tohoto projektu spočívá v překlonování katalytické domény proteinu Alg1 do jiného vektoru, např. do vektoru pMALc2ErsTEV. Tento vektor přidává na N-terminální konec proteinu maltosu-vázající protein (MBP), a tím zvyšuje rozpustnost proteinu a umožňuje purifikaci.

Abstract:

The presented thesis is focused on study of glycosyltransferases, specifically on the β-1,4-mannosyltransferase from the human pathogen A. fumigatus. GTs are significantly involved in the synthesis of the cell walls of pathogenic organisms, and thus their virulence. Therefore, knowledge of these enzymes is very important. Preparation of GTs is very difficult, because they are very often transmembrane proteins and it is necessary to optimize conditions for their preparation. The project was focused β-1,4-mannosyltransferase (Alg1) and its catalytic domain. Gene for the enzyme and its shorted part were cloned into the pET29b vector for expression in E. coli host cells. Preparation of the whole protein was not successful, even using different expression strains and conditions. After optimization, the catalytic domain of the protein was produced in the E. coli BL21 (DE3) cells. Unfortunately, the most of the protein was produced in insoluble inclusion bodies, therefore purification of the Alg1 catalytic domain under denaturing conditions was tried. Purification under native conditions was also tested, because a small amount of the protein was found in the soluble component. Three different resins for the purification using immobilized metal ion affinity chromatography were tested – Ni-NTA, Cu-NTA and Cu-IDA. None of them resulted in successful purification. As the protein did not bind to the column. However, the presence of the oligohistidine anchor in the protein was confirmed by mass spectrometry. In the future the project should continue by cloning of the Alg1 protein catalytic domain into another vector, such as the vector pMALc2ErsTEV. This vector adds the Maltose-Binding Protein (MBP) to the N-terminal part of proteins, which increases the solubility of proteins and facilitates their purification.
