Abstract:

Habilitační práce se zabývá fyziologickými reakcemi lišejníků na změny fyzikálních faktorů prostředí. Práce je zaměřena na přehled reakcí různých druhů lišejníků, především z polár-ních oblastí, jejichž stélky byly experimentálně vystaveny chladovým a mrazovým teplotám, a/nebo různému stupni vysychání (dehydratace). Fyziologické procesy extremofilních autotrofních organismů (lišejníky, řasy, mechy) z polár-ních oblastí Země byly v posledním desetiletí velmi intenzivně studovány. Umožňují organis-mům úspěšně přežívat a reprodukovat se v extrémních podmínkách polárních a vysoko-horských oblastí. Tyto organismy využívají množství biofyzikálních, biochemických i moleku-lárně biologických mechanismů, které se aktivují během působení chladu a mrazu, nedo-statku vody, cyklů vysychání (dehydratace) a zavodňování (hydratace), střídání polárního dne a noci, působení silného UV záření, nebo měnícího se množství a spektrálního složení dopadajícího slunečního záření. V první části habilitační práce jsou stručně shrnuty nejnovější poznatky o vybraných as-pektech vlivu chladového a mrazového stresu na rostliny a lišejníky. Pozornost je věnována zejména (1) mechanismům tolerance stélek lišejníků k chladu a mrazu, (2) teplotnímu optimu fotosyntézy lišejníků, (3) fotosyntetické aktivitě stélek za teplot pod 0°C, a (4) roli nukleace ledu (tvorba krystalizačních zárodků ledu), respektive intracelulárnímu a extracelulárnímu mrznutí vody ve stélkách lišejníků. Součástí práce je i přehled aktuálních poznatků o foto-syntéze lišejníků za různého stupně hydratace stélek, včetně popisu mechanismů tolerance lišejníků k dehydrataci, a jejímu vlivu na anatomii a fyziologii lišejníkové stélky. Ve druhé části práce jsou stručně popsány metody využívané při mé experimentální práci a hlavní zjištěné výsledky vybraných publikací. Mezi nejvýznamnější poznatky těchto prací patří: (1) stimulační (pozitivní) účinek ribitolu na fotosyntetické procesy lišejníků a řas za nízkých teplot, (2) druhově specifické rozdíly v kritické teplotě pro primární procesy foto-syntézy vybraných druhů lišejníků, (3) vysoká odolnost stélek lišejníků vůči extrémně nízkým teplotám v závislosti na rychlosti jejich zmražení, (4) výrazné mezidruhové rozdíly v senziti-vitě k zmrznutí u lišejníkových symbiotických řas rodu Trebouxia, (5) druhově-specifický trend v poklesu signálu fluorescence chlorofylu v průběhu vysychání stélek lišejníků (indi-kující limitaci primárních procesů fotosyntézy) a (6) dehydratací-indukované změny v in-dexech spektrální reflektance a ve fluorescenci chlorofylu. Součástí habilitační práce jsou přiložené impaktované odborné publikace autora.

Abstract:

The habilitation thesis deals with physiological lichen responses to the changes in physical factors of the environment. It is focused on the responses of several lichen species, mostly collected in polar regions, exposed to the experimental cold and freezing temperature and/or dehydration of their thalli. Extremophile autotrophs (such as lichens, algae, mosses) in Polar Regions have been intensively studied within last decade. Their physiological processes enable them to survive and reproduce themselves in extreme conditions of polar and alpine regions. Extremophile autotrophs use a great number of different biophysical, biochemical and molecular mechanisms that are activated while exposed to different stresses, such as cold and freezing temperature, dehydration, and hydration-dehydration cycles, changes of polar day and night, high UV radiation, or changes in quantity and spectral composition of incident irradiance. The first part of the thesis is a review of the most recent knowledge about selected aspects of the effects of cold and freezing stress on plants and lichens. It is focused primarily on (1) mechanisms of cold and freezing stress tolerance in lichen thalli, (2) temperature optimum for photosynthetic processes in lichens, (3) photosynthetic activity of lichen thalli in freezing temperature (below 0°C), and (4) the ice nucleation, intracellular and extracellular ice formation in lichen thalli. The habilitation thesis also includes a review of latest findings on lichen photosynthesis as affected by different levels of dehydration, description of lichen dehydration-tolerance mechanisms as well as the effect of dehydration on lichen anatomy and physiology. Second part of the thesis consists of a brief review and characterization of the methods used in my experimental work and summary of the most important results of selected publications related to the main topic. The most important highlights include: (1) positive effect of ribitol on photosynthetic processes of lichens and algae affected by low temperature, (2) species-specific differences in critical temperature for primary processes of photosynthesis in selected lichen species, (3) high resistance of lichen thalli to extremely low temperature in relation to the cooling rate, (4) significant interspecies differences in freezing sensitivity of lichen symbiotic alga Trebouxia, (5) species-specific decrease of chlorophyll fluorescence parameters during lichen thalli dehydration, indicating limitation of primary photosynthetic processes, and (6) dehydration-induced changes in spectral reflectance indices and chlorophyll fluorescence. The author’s most important publications in impacted journals are attached to the thesis.
