Epigenetic regulation of floral genes

Abstract:

Cílem bakalářské práce „Epigenetická regulace genů kvetení“ je prostudovat vliv hypometylačních činidel dihydroxypropyladeninu a zebularinu na transkripci květních genů AP1, PI a genu PHE1 majícího vliv na vývoj semen u modelové rostliny Arabidopsis thaliana. Teoretická část práce se věnuje epigenetickým modifikacím, především metylaci cytosinu. Dále jsou charakterizovány analyzované geny. V praktické části jsme zjistili, že přítomnost hypometylačních látek během klíčení semen nevedla ke zvýšení transkripce genů AP1 a PI v pupenech A. thaliana, působení zebularinu vedlo dokonce k poklesu transkripce analyzovaných genů, přičemž efekt byl výraznější u genu AP1. Analýza transkripce genu PHE1 nebyla provedena, protože přes veškerou optimalizaci izolačních postupů RNA získaná ze šešulí nebyla dostatečné kvality. V diskusi jsou porovnány výsledky získané v této studii s analogickým experimentem na modelové rostlině Nicotiana tabacum.

Abstract:

The aim of this bachelor thesis „Epigenetic regulation of floral genes“ is to study the effect of hypomethylating drugs dihydroxypropyladenine and zebularine on the transcription of floral genes AP1, PI and of PHE1 gene which is involved in seeds development in a model plant Arabidopsis thaliana. In the theoretical part, literature data about DNA methylation are presented, and the analyzed genes are described. In the practical part, transcription of selected genes in tissues of plants germinated in the presence of hypomethylation drugs is analyzed. According to our results, treatment with hypomethylating drugs during seeds germination did not increase the transcription of AP1 and PI genes in A. thaliana’s buds. Surprisingly, transcription of analyzed genes was even lower in zebularine - treated individuals and this effect was more pronounced in case of AP1 gene. Analysis of PHE1 gene was not accomplished, because isolation of RNA from siliques represents a serious methodical problem and despite our effort we were not able to get samples of required quality. Results are discussed with analogous experiment using another model plant Nicotiana tabacum.
