Analysis of stress response in non-target organisms after protective sprays

Abstract:

V posledních letech se rozšiřuje problém, kterým je negativní účinek ochranných postřiků na necílové organismy. Přestože jsou ochranné postřiky pro zemědělství užitečné, tak poškozují například opylovače, kteří jsou nepostradatelní pro růst jakýchkoliv rostlin. Tato bakalářská práce obsahuje shrnutí informací o stresové odpovědi necílových organismů na příkladu včely medonosné (Apis mellifera) a čmeláka zemního (Bombus terrestris L.) po expozici pesticidům. Změny genové exprese byly studovány pomocí toxikogenomických metod. Použité metody byly konkrétně RNA sekvenování (RNA-seq) a kvantitativní polymerázová řetězová reakce (qRT-PCR). Pomocí těchto metod jsou odhaleny negativní účinky pesticidů, díky kterým se určí, jestli je vhodné daný postřik používat či nikoli.

Abstract:

In the last few years, the negative effect of protective sprays on non-target organisms has become a growing problem. Although protective sprays are useful for agriculture, they represent harm, for example to pollinators, which are indispensable for the growth of any plant. This thesis summarises information about the stress response of non-target organisms using the honey bee (Apis mellifera) and the bumblebee (Bombus terrestris L.) as examples, following exposure to pesticides. Changes in gene expression were studied by using toxicogenomic methods. The used methods were RNA sequencing (RNA-seq) and quantitative polymerase chain reaction (qRT-PCR). These methods are used to detect the negative effects of pesticides and determine whether or not a given spray is appropriate.
