Detection of DNA damage by means of electrochemical methods.

Abstract:

Poškození DNA může mít za následek vznik mnoha vážných chorob. V poslední době je poškození DNA věnována velká pozornost a hledají se co nejjednodušší a zároveň co nejúčinnější způsoby detekce tohoto poškození a látek, které mohou poškození DNA způsobit. Elektrochemické metody by měly představovat rychlý a účinný způsob detekce poškození DNA. Tato práce byla zaměřena na detekci jednořetězcových zlomů pomocí AC voltametrie. Cílem bylo srovnání citlivosti elektroforetických a elektrochemických metod. V teorii je stručné shrnutí elektrochemických vlastností nukleových kyselin a působení použitých genotoxických látek. K modifikaci DNA byl použit manganistan draselný a hydroxylové radikály, jejichž účinky na DNA byly již poměrně dobře popsány.

Abstract:

Damage to DNA may result in generation of serious diseases. In recent years, DNA damage has been intensively studied. The biggest effort was spent on finding of the most effective and the simplest detection method of DNA damage and DNA-damaging agents. Electrochemical methods should be fast and effective way of the detection. Main goal of this thesis was detection of DNA single strand breaks by means of electrochemical methods. In theoretical part, there are briefly discussed electrochemical properties of nucleic acids and effect of used genotoxic agents. KMnO4 and hydroxyl radicals were used for DNA modification, their effects on DNA were quite well-described.
