Vliv hypometylace DNA, vernalizace a genů pozdního kvetení na expresi genu FLC

Abstract:

Hlavním cílem této práce byla analýza vlivu genů M73, L4 a Spi na expresi genu FLC u Arabidopsis thaliana. Gen M73 byl na základě demetylačních a vernalizačních pokusů zařazen do autonomní dráhy (Lízal a Relichová, 2001), která má na expresi genu FLC inhibiční vliv. Jak ukázaly expresní analýzy genu FLC, u linie M73 tomu není jinak. Touto prací jsem tak potvrdila dosavadní výsledky regulace u autonomní dráhy. U linií L4 a Spi, zařazených do dosud neanalyzované dráhy metylační (Lízal a Relichová, 2001) jsem zjistila, že tyto geny mají na expresi genu FLC také inhibiční vliv. U této dráhy se jedná o zcela nový poznatek. V další části této práce byl stanovován expresní profil genu FLC po působení vernalizace a 5-azacytidinu. Překvapivě zde nedocházelo ani u jednoho z analyzovaných mutantů k poklesu exprese inhibitoru kvetení FLC po těchto působení a to ani v době květu. Toto přetrvávání exprese může být způsobeno přemostěním působení genu FLC jiným genem například SOC1 nebo přímým působení na geny řídící tvorbu květních meristému LFY nebo AP1. Vzhledem k tomu, že výsledky expresních analýz úplně neodpovídaly dosavadním poznatkům, analyzovala jsem v poslední části této práce delece v oblastech s dopadem na regulaci exprese FLC. Molekulární analýzou funkčních oblastí tohoto genu jsem prokázala četné delece v oblasti promotoru, prvních dvou exonů a prvního intronu. A to v regulačních oblastech podílejících se na udržení represe genu FLC po vernalizaci, ale také v oblasti zajišťující negativní regulaci tohoto inhibitoru u nevernalizovaných rostlin. U této části práce se však zatím jednalo o pilotní studii, která bude vyžadovat ještě další podrobnější zkoumání.

Abstract:

The main objective of this thesis was an analysis of the influence of genes M73, L4 and Spi on expression of FLC gene in Arabidopsis thaliana. Based on experiments with vernalization and demethylation M73 gene was put into the autonomous regulation pathway (Lízal and Relichova, 2001). that repress FLC gene. As the expression analyses showed, M73 gene has the same infulence. So, in this thesis I have contemporary results of autonomous pathway regulation. Analysing mutants L4 and Spi imbedded in a still not explored methylation pathway (Lizal and Relichova, 2001) I have learnt that these genes also repress FLC. That means an entirely new information in this regulation pathway. In the following part of this thesis an expression pattern after vernalization a demethylation was made. Suprisingly, none of the analysed mutants showed a decrease of inhibitor of flowering FLC after this treatment neither in flowering time. It s possible that continual expression could be overcome by another gene e.i. SOC1 or by direct incidence on flower-meristem/organ identity genes such as LFY or AP1. Because the results of expression analysis did not fully corespond with the present results, in the last part of this thesis I analysed deletions in areas responsible for regulation of FLC expression. I demonstrated some deletions in promotor, the first two exons and the first intron. These deletions were in regulation areas that share control of repression of FLC after vernalization but also in the area negatively regulating expression of this inhibitor in nonvernalized plants. However, this part of the thesis was just a pilot project that will demand more detailed further studies.
