The role of AGR2 in the epithelial-mesenchymal transition.

Abstract:

Anterior gradient 2 (AGR2) je protein, jehož zvýšená exprese je spojená s řadou nádorových onemocnění. Mezi hlavní funkce AGR2 patří regulace přežívaní nádorových buněk, regulace buněčné proliferace, tvorba rezistence k protinádorovým léčivům a aktivně se podílí na vzniku metastáz, avšak přesný mechanizmus není objasněn. Cílem této práce byla analýza úlohy proteinu AGR2 v procesu epiteliálně-mezenchymální tranzice (EMT), v procese, který představuje klíčový krok při tvorbě metastáz. Pomocí kvantitativní PCR a imunochemické analýzy jsme zjistili, že EMT indukovaná pomocí TGF-β, způsobuje pokles hladiny AGR2, a to na úrovni mRNA i proteinů na panelu různých buněčných linií. Vliv TGF-β byl ověřen i mikroskopicky pomocí imunofluorescenční detekce vybraných proteinů. Extrémně zajímavým bylo zjištění, že umlčením AGR2 exprese pomoci specifických siRNA vedlo ke zvýšení hladiny mezenchymálnich markerů a naopak redukci hladiny epiteliálních determinant. Při opačném postupu, kdy byl vnesen AGR2 plasmid do AGR2 negativních buněk byla pozorována indukce cytokeratinu 18 a snížení hladiny mezenchymálnich markerů. Při testování vlivu hypoxie na indukci EMT, jsme prokázali, že hypoxické podmínky vedou ke snížení hladiny AGR2 a zvýšeni exprese vimentinu. Naše výsledky jasně potvrzují, že AGR2 je negativně regulovaná TGF-β, přičemž exprese AGR2 souvisí nejen s epiteliálnym fenotypem ale pravděpodobně je i přímo zapojena do regulace EMT.

Abstract:

The anterior gradient 2 (AGR2) is a protein overexpressed in many type of cancers. AGR2 was shown to be involved in regulation of cancer cell survival, cellular proliferation, development drug resistance and formation of metastasis; however the exact mechanism is still not understood. Since the epithelial-mesenchymal transition (EMT) represents a key step in the tumor progression leading to metastasis development, the aim of this work is to analyze the role of AGR2 in this process. Quantitative PCR and immunochemical analysis showed that EMT induced by TGF-β1 causes the decrease of AGR2 at both mRNA and protein level in panel of different cell lines. The effect of TGF-β was also verified using microscopy by immunofluorescence detection of selected proteins. Interestingly, we also found that siRNA-mediated AGR2 knockdown induces upregulation of mesenchymal marker vimentin and the decrease of epithelial E-cadherin. In addition, introduction of plasmid with AGR2 into the AGR2 negative cell lines induced expression of cytokeratin 18 and attenuated expression of mesenchymal markers. Moreover, we tested the effect of hypoxia on the induction of EMT and found that hypoxic conditions correlate with partial loss of AGR2 and increased expression of vimentin. Our findings confirmed that AGR2 is negatively regulated by TGF-β and that AGR2 expression is associated not only with epithelial phenotype but would be directly involved in regulation of EMT.
