Use of DNA markers to evaluate the potential of grapevine genotypes for breeding to resistance to fungal diseases

Abstract:

Tato diplomová práce se zabývá Rpv a Run/Ren lokusy, díky nimž se réva vinná aktivně brání při napadení houbovými patogeny. Literární přehled je zaměřen na šlechtění révy vinné na rezistenci vůči houbovým chorobám. Jsou zde popsány houbové patogeny P. viticola Berl. & De Toni a U. necator Schwein a způsob jakým napadají révový keř. Následuje popis současných SSR metod využívaných při genetickém mapování odrůd. Velká část je věnována vybraným Rpv a Ren lokusům, se zaměřením na jejich původ, umístění v genomu révy a míru poskytované odolnosti. Následuje popis spolupráce jednotlivých lokusů mezi sebou a vysvětlením pojmu pyramidizace genů rezistence. V experimentální části byl zvolen soubor odrůd u nichž se provedla izolace genomické DNA, která byla poté využita pro analýzu všech kultivarů pomocí SSR markerů. Získané amplikony byly vyhodnoceny pomocí genetického analyzátoru a následovalo posouzení přítomnosti či absence rezistentního lokusu.

Abstract:

This dissertation deals with the issue of Rpv and Run/Ren loci, which enable grapevine to actively defend against fungal pathogens. The introductory chapter presents a literature review focused on grapevine breeding for mildew resistance, specifically concentrating on the P. viticola Berl. & De Toni a U. necator Schwein and the way, they attack the grapevine bush. The next chapter thoroughly describes the contemporary SSR methods used for the genetic mapping of species. Substantial attention is devoted to the selected Rpv and Ren loci, primarily focusing on their origin, location in the grapevine genome, and the extent of resistance provided. The next chapter deals with describing the cooperation of individual loci and the explanation of the concept of pyramiding resistance genes. The experimental part presents a set of species in which the isolation of genomic DNA was accomplished. This DNA was used for the analysis of all cultivars using SSR markers. The obtained amplicons were assessed using a genetic analyzer, which was followed by the evaluation of the presence or absence of a resistant locus.
