Role of Insulin in the Brain Tissue

Abstract:

Inzulin, produkovaný slinivkou břišní, slouží především jako signál pro transport glukózy do těch buněk těla, které jsou na něm závislé. Inzulinové receptory jsou však exprimovány v celém mozku, i když tyto buňky inzulin k transportu glukózy nepotřebují. Tato práce je zaměřena na sumarizaci poznatků o funkci inzulinu v mozku. Inzulin je transportován přes hematoencefalickou bariéru a váže se na receptory v CNS a řídí tak periferní metabolizmus. Inzulin působící v CNS ovlivňuje totiž řadu metabolických funkcí počínaje chutí k jídlu, reprodukci, tělesnou teplotu, mitochondriální funkce, metabolizmus glukózy a další. Inzulinová signalizace také ovlivňuje nálady, učení, paměť a s ní spojenou synaptogenezi. Rezistence na centrální inzulin přispívá k depresím a rozvoji neurodegenerativních onemocnění, zejména Alzheimerovy choroby. Tato bakalářská práce dokazuje, že i přes relativní novost a s tím spojenou nedokonalou znalost výše uvedených mechanizmů je zjevné, že funkce inzulinu v mozku je extrémně důležitá pro homeostázu celého organizmu.

Abstract:

Insulin, produced by the pancreas, is primarily used as a signal for the transport of glucose into the insulin-dependent cells of a body. However, insulin receptors are expressed throughout the brain, although the glucose transport to these cells is not insulin-dependent. This work is focused on summarizing findings about the function of insulin in the brain. Insulin is transported through the blood-brain barrier, and binds to receptors in the CNS and controls the peripheral metabolism. Insulin acting in the CNS affects many metabolic functions as appetite, reproduction, body temperature, mitochondrial function, glucose metabolism, etc. Insulin signaling also influences mood, learning, memory and associated synaptogenesis. Resistance to central insulin contributes to the development of depression and neurodegenerative diseases, particularly Alzheimer's disease. This thesis shows that despite the relative novelty and thus imperfect knowledge of the mechanisms described above, it is obvious that the function of insulin in the brain is extremely important for the homeostasis of the whole organism.
