Characterization of the binding specificity of the plant telomeric sequence binding proteins

Abstract:

Telomerické repetice se nejčastěji vyskytují na fyzických koncích eukaryotických chromozomů, ale mohou být lokalizované i v jiných částech rostlinného genomu. V mnoha organizmech za vazbu proteinů na dvouvláknovou telomerickou DNA zodpovídá Myb-like doména typu telobox. Rostlinné proteiny s Myb-doménou typu telobox na svém N-konci jsou řazeny do rodiny SMH (Single Myb Histone). Členové této rodiny, proteiny AtTRB (Telomere Repeat Binding), jsou schopni vazby na rostlinné telomerické repetice in vitro, avšak přesná lokalizace těchto proteinů v rámci rostlinného genomu zůstává stále nejasná. Optimalizací chromatinové imunoprecipitace a následnou hybridizací s telomerickou sekvencí jsme prokázali, že protein AtTRB1 je asociován s telomerickou sekvencí in vivo. Zároveň byla lokalizace proteinu AtTRB1 pozorována pomocí imunofluorescenčních metod v jádru a jadérku. Analýzou TRF bylo prokázáno zkracovaní telomer u rostlin mutantních pro protein AtTRB1, avšak změna aktivity telomerázy in vitro pozorována nebyla.

Abstract:

Telomeric repeats most often occur at the physical ends of the eukaryotic chromosomes, but may be located in other parts of the plant genome. The telobox, a Myb-related telomeric DNA binding motif is responsible for protein binding to double-stranded telomeric DNA in many organisms. Plant proteins with telobox at its N-terminus are classified into the family SMH (Single Myb Histone). Members of this family, proteins AtTRB (Telomere Repeat Binding) are capable of binding to plant telomeric sequence in vitro, but the precise localization of these proteins in the plant genome remains unclear. Optimization of chromatin immunoprecipitation and subsequent hybridization with telomeric sequences demonstrated that the protein AtTRB1 is associate with telomeric sequences in vivo. Also AtTRB1 protein localization in the nucleus and nucleolus was observed by imunofluorescence methods. TRF analysis showed shortening of telomeres in plants that are null mutants for AtTRB1, although changes were not observed in telomerase activity in vitro.
