Pharmaceuticals as new emerging contaminants in aquatic environment and their effects on non-target organisms

Abstract:

Farmaka se vyskytují v prostředí díky svým vlastnostem nejvíce v povrchových a podzemních vodách, ale také v půdách a sedimentech. Hlavními zdroji jsou humánní a veterinární použití farmak, po kterém vstupují do životního prostředí s odpadní vodou, která může být s různou úspěšností čištěna, nebo hnojením polí pomocí odpadního kalu nebo hnoje. Díky tomu, že jsou farmaka navrhována tak, aby měla biologický efekt, mají po svém vstupu do prostředí stále velký potenciál k ovlivnění necílových organismů. Bylo už prokázáno, že farmaka způsobují efekty v životním prostředí. Cíle této práce byly sesbírat a studovat současnou literaturu věnující se problematice farmak v prostředí, a to zvláště jejich efektům a také jejich výskytu v prostředí. Dále vybrat farmaka, která budou testována v rámci práce a následně je otestovat na baterii akutních ekotoxikologických biotestů, která obsahuje testy s řasami a sinicemi, vodním korýšem a bakteriemi. S pomocí výsledků také zhodnotit riziko, které mohou farmaka v životním prostředí mít. Tato práce je rozdělena na teoretickou a praktickou část. Teoretická část obsahuje shrnutí informací o výskytu farmak v prostředí a také o jejich osudu v prostředí a jejich vlastnostech. V praktické části jsou prezentována získaná data a jsou diskutována s dostupnou literaturou. Předpokládané koncentrace bez efektu, vypočítané z výsledků této práce, byly porovnány s koncentracemi v prostředí. Farmaka vybraná pro testování byla ciprofloxacin, diklofenak, hydrochlorothiazid, ibuprofen, karbamazepin, klofibrová kyselina, paracetamol a sulfamethoxazol. Vybraná farmaka jsou ze skupin analgetik, antiepileptik, hypolipidemik, diuretik a antibiotik. Baterie akutních biotestů obsahuje test inhibice růstu řas a sinic, test akutní imobilizace Daphnia magna, Microtox a SOS Chromotest. Z testovaných farmak měl nejtoxičtější efekty ciprofloxacin pro sinici Microcystis aeruginosa. Nejcitlivějším testem v baterii byl Microtox a nižší toxicita byla hodnocena pomocí řasového a dafniového testu. Jediný ciprofloxacin byl prokazatelně genotoxický.

Abstract:

Pharmaceuticals are present in the environment due to their properties mostly in surface and ground water but also in soils and sediments. The main sources are the human and veterinary therapeutic usage, after which they enter the environment with waste water, which can be more or less treated, or by dispersion of sludge or manure on soil. As pharmaceuticals are designed to have biological effects, they are unfortunately still able to affect the non-target organisms in the environment as they enter it. There are some environmental effects linked solely to the pharmaceuticals (Kidd et al., 2007; Oaks et al., 2004). Aims of this thesis were: to collect and summarize the present literature on pharmaceutical effects and occurrence in the environment; select the pharmaceutical for ecotoxicological testing and subject them to the battery of acute test with algae and cyanobacteria, crustaceans and bacteria; and to evaluate the risk they might have in the environment . This thesis is divided into theoretical and practical part. In theoretical part, the information about pharmaceuticals occurrence, fate and properties in the environment are given. In the practical part, the acquired results and discusses are presented and discussed with literature findings. Predicted no-effect concentrations were calculated and compared with environmental concentrations to estimate the environmental risk. The pharmaceuticals chosen for testing were carbamazepine, ciprofloxacin, clofibric acid, diclofenac, hydrochlorothiazide, ibuprofen, paracetamol, and sulfamethoxazole. They represent the groups of NSAIDs, anti-epileptics, hypo lipidemics, diuretics and antibiotics. The battery of acute ecotoxicological tests consists of algae and cyanobacteria growth inhibition test, acute immobilization test with Daphnia magna, Microtox and SOS Chromotest. From the tested substances, ciprofloxacin had the most toxic effect on cyanobacteria. Most sensitive from the tests was Microtox and daphnia and algae tests have the highest effective concentrations. The only pharmaceutical with genotoxic properties was ciprofloxacin.
