Occurrence and distribution of metazoan parasites within communities of host fih

Abstract:

Předkládaná disertační práce se skládá ze tří částí. Studovány byly sezonní změny v početnosti, prostorová distribuce a hostitelská specifita parazitů sladkovodních ryb v souvislostech s faktory hostitele a prostředí. První část studie se zabývá výskytem a prostorovou distribucí glochidií, larválních stadií sladkovodních mlžů čeledě Unionidae, parazitujících převážně na ploutvích a žábrách sladkovodních ryb. Celkem bylo na přítomnost glochidií vyšetřeno 22 druhů tří čeledí ryb. Distribuce glochidií na hostiteli byla podrobně studována na třech běžných druzích ryb plotici obecné (Rutilus rutilus (L.)), okounu říčním (Perca fluviatilis L.) a hořavce duhové (Rhodeus sericeus (Pallas)). Ryby byly loveny v průběhu tří let v oblasti aluvia řek Morava a Kyjovka (Česká republika). Zjištěna byla glochidia dvou rodů, Anodonta a Unio. Glochidia rodu Anodonta byla nalezena na 10 druzích a glochidia rodu Unio na 17 druzích ryb. Glochidia rodu Unio byla lokalizována převážně na žábrách, kdežto většina glochidií rodu Anodonta na ploutvích ryb, nejvíce na okrajích prsních ploutví. Glochidia rodu Unio byla na žábrách zachycena nejčastěji na druhém a třetím žaberním oblouku. Byla potvrzena sezonnost výskytu glochidií. V průběhu zimy a na jaře byla pozorována glochidia rodu Anodonta, kdežto glochidia Unio byla nejpočetnější od května do června. Ze sezonně sledovaných druhů ryb byly nejvyšší abundance glochidií zachyceny na okounovi, nižších hodnot dosahovali na plotici. I přes blízkou vazbu hořavky a mlže byla glochidióza na tomto druhu ryby vzácná. V případě vysokých intenzit infekce glochidií u okouna říčního byl potvrzen pozitivní vztah mezi počtem glochidií a délkou ryb. V druhé části předkládané práce byl studován vliv sezonních změn na složení parazitárního společenstva a prostorová distribuce monogeneí parazitujících na hrouzku obecném (Gobio gobio (L.)) v průběhu dvou let na řece Hané (Česká republika). Nalezeno bylo 6545 jedinců parazitů (8 druhů monogeneí a 3 druhů digeneí). Parazitárnímu společenstvu dominovali zástupci třídy Monogenea a zaznamenány byly druhy: Dactylogyrus cryptomeres, Gyrodactylus gobiensis, G. gobii, G. gasterostei, G. vimbi, G. markakulensis, G. sedelnikowi a Paradiplozoon homoion. Metacerkárie digeneí byly nacházeny v malé míře; Rhipidocotyle illense, Rossicotrema donicum a Tylodelphys clavata. V roce 2001 dosáhli zástupci rodu Gyrodactylus maximálních hodnot abundance v březnu a D. cryptomeres o dva měsíce později, v květnu. V roce 2002, kdy byla teplota vody v průměru nižší, dosáhli gyrodaktylové maximální abundance v květnu a D. cryptomeres v červenci. Většina jedinců rodu Gyrodactylus byla nalezena na ploutvích (70-95%), zbývající jedinci na povrchu těla a žábrách ryb, kde se nacházeli všichni jedinci rodu Dactylogyrus a Paradiplozoon. Preference mikrohabitatů monogeneí se lišila v závislosti na teplotě vody a intenzitě infekce. Ve třetí, experimentální části disertační práce byly příbuzné druhy rybích hostitelů chovány ve skupinách samostatně nebo společně. V rámci těchto skupin byla sledována schopnost monogeneí a zejména zástupců rodu Gyrodactylus měnit hostitele. První skupina hostitelů zahrnovala okounovité ryby, okouna říčního (P. fluviatilis) a ježdíka obecného (Gymnocephalus cernuus (L.)), druhá skupina zahrnovala ryby kaprovité, plotici obecnou (R.. rutilus) a střevli potoční (Phoxinus phoxinus (L.)). Celkem bylo na experimentálních rybách pozorováno 18 druhů monogeneí 5 rodů (Ancyrocephalus, Dactylogyrus, Gyrodactylus, Paradiplozoon, Pellucidhaptor), z čehož bylo determinováno 8 druhů rodu Gyrodactylus. k se jevil být vhodnějším hostitelem pro tento druh parazita. Na plotici byly nalezeny 3 druhy gyrodaktylů a na střevli 6 druhů. Pouze G. vimbi, jež parazitoval oba druhy ryb, zaznamenal nárůst početnosti v experimentálních podmínkách na obou hostitelích. Druhy Gyrodactylus rutilensis na plotici a Gyrodactylus pannonicus na střevli se jevili jako specialisti, jelikož ve společných nádržích hostitele neměnily. Gyrodactylus macronychus byl jediným druhem, který prokazatelně změnil hostitele, a to ze střevle na plotici. Většina gyrodaktylů na střevli a plotici byla nalezena na ploutvích ryb, kdežto na ježdíku a okounovi parazitovali gyrodaktylové převážně na hlavě a žaberním aparátu.

Abstract:

The PhD thesis consists of three parts. Seasonal changes in occurrence, microhabitat selection and host specificity of metazoan parasites of freshwater fish in relation to biotic and abiotic factors were studied. In the first part of the thesis, occurrence and microhabitat preference of glochidia (the larval stages of freshwater unionid mussels) parasitizing the fins and gill apparatus of fish was studied. A total of 22 fish species were examined for the presence of glochidia and their distribution on individual hosts was studied on 3 common fish species: roach (Rutilus rutilus (L.)), perch (Perca fluviatilis L.) and bitterling (Rhodeus sericeus (Pallas)). The fish were obtained from the floodplain of the Morava and Kyjovka Rivers over a period of three years. The glochidia of two genera, Unio and Anodonta, were found. The Anodonta glochidia were observed on 10 fish species and Unio glochidia on 17 fish species. There was a significant difference in microhabitat distribution of glochidia on the body of the host fish. Unio glochidia were predominantly located on the gills, whereas most Anodonta glochidia were found on the fins, with the highest numbers of glochidia observed on the margin of the pectoral fins. For the gill apparatus, Unio glochidia were found predominantly on the second and third arch. Anodonta glochidia were predominantly found during winter and spring (November – May), whereas Unio glochidia were more abundant during May and June. The number of glochidia positively correlated with fish length on perch highly infected by Anodonta glochidia and all perch infected by Unio glochidia. Of the three seasonally studied fish species, the highest occurrence of parasites was found on perch with fewer numbers observed on roach. In spite of the close relationship between bitterling and unionid mussels, glochidiosis was rare on this fish species. In the second part of the thesis, seasonal changes in epidemiology, parasite species composition and microhabitat preference of the monogenean parasites of gudgeon (Gobio gobio (L.)) were studied during two years in the River Haná (Czech Republic). Altogether 230 specimens of host fish were examined for the presence of metazoan parasites, and a total of 6545 specimens of eight monogenean and three digenean species were recorded. Monogeneans dominated the parasite community and following species were found: Dactylogyrus cryptomeres, Gyrodactylus gobiensis, G. gobii, G. gasterostei, G. vimbi, G. markakulensis, G. sedelnikowi and Paradiplozoon homoion. Each specimen of three digenean species was found as metacercarie stage. Digenean species found were: Tylodelphys clavata, Rossicotrema donicum and Rhipidocotyle illense. In 2001, the peak of gyrodactylid infection was recorded in March and D. cryptomeres infection peaked two months later in May. In 2002, gyrodactylid abundance peaked in May and D. cryptomeres in July. Majority of Gyrodactylus specimens were found on the fins (70-95%); remaining specimens were located on the body surface and the gills, where all Dactylogyrus and Paradiplozoon homoion specimens were located. Microhabitat preference of monogeneans changed in accordance to the water temperature and intensity of infection. In the third, experimental part of the thesis, related fish species were kept in single- or mixed-species groups to test host specificity and the ability of Gyrodactylus species to change their host fish under experimental conditions. The first host group included two percid fish species, perch (P. fluviatilis) and ruffe (Gymnocephalus cernuus (L.)), the second host group included two cyprinid species, roach (R. rutilus) and minnow (Phoxinus phoxinus (L.)). A total of 4182 specimens of 19 monogenean species belonging to five genera were observed, with 8 Gyrodactylus species identified. f Gyrodactylus found on roach and six species on minnow. Gyrodactylus vimbi was the only species that seemed to increase in abundance during the experiments. Gyrodactylus vimbi parasitized both cyprinid fish species. Gyrodactylus rutilensis and Gyrodactylus pannonicus did not change the host fish species in the mixed species tanks and seemed to be specialists for roach and minnow, respectively. Gyrodactylus macronychus was the only species that apparently changed hosts; from minnow to roach. Most of the gyrodactylids observed on roach and minnow were located on the fins, while gyrodactylids of perch and ruffe parasitized mainly the gill chamber and head.
