Carboxypeptidase B1 a possibilities of its therapeutical modulation

Abstract:

Tato diplomová práce je zaměřena na karboxypeptidasu B1 (CPB1), protein typicky spojený s pankreatitidou, zvýšené hladiny tohoto proteinu byly však detekovány také u nádorů prsu. Na základě předchozí proteomické a transkriptomické analýzy byla karboxypeptidasa B1 identifikována jako potenciální biomarker metastazování do lymfatických uzlin u nádorů prsu subtypu luminální A. Prvním cílem této práce bylo analyzovat vliv exprese CPB1 na vlastnosti nádorových buněk, především tuhosti, která byla analyzována pomocí mikroskopie atomárních sil. Bylo zjištěno, že zvýšená hladina CPB1 způsobuje významné snížení tuhosti nádorových buněk MCF-7, zároveň byly tyto buňky také morfologicky odlišné od buněk neexprimujících protein CPB1. Dále v této práci byl ověřen efekt panelu potenciálních antimetastatických inhibitorů na hladinu CPB1, a to za účelem charakterizovat mechanismus prometastatického působení CPB1 a nastínit možné cesty jejího terapeutického ovlivnění. Hladinu CPB1 významně ovlivnily dva inhibitory. Inhibitor norcantharidin hladinu CPB1 významně snižoval po 24 hodinové inkubaci. Naopak inhibitor ATM proteinu, KU 60019, hladinu CPB1 zvýšil po 24 hodinovém působení. Oba tyto inhibitory by měly vést buňky do apoptózy, což bylo testováno pomocí průtokové cytometrie. Působení inhibitoru norcantharidin buňky MCF-7 CPB1 vedlo do apoptózy a pozdní apoptózy. Naopak bylo zjištěno, že inhibitor KU 60019 způsoboval nekrózu buněk linie MCF-7 CPB1, nikoliv jejich apoptózu. Pomocí LC-MS/MS v „data independent acquisition“ (DIA) módu byl porovnán proteom buněk MCF-7 CPB1 s proteomem kontrolní linie MCF-7 GFP. U linie MCF-7 CPB1 byla zvýšená hladina markerů pro epiteliálně-mezenchymální tranzici, dále zvýšená hladina proteinů podporující agresivitu nádorových buněk a také angiogenezi, sníženy byly hladiny proteinů zajišťující buněčnou adhezi. Tato data poukazují na možný vztah CPB1 k epiteliálně-mezenchymální tranzici a naznačují možné způsoby modulace jeho hladiny.

Abstract:

In this thesis we study carboxypeptidase B1 (CPB1), a protein associated with pancreatitis. Increased levels of CPB1 were also detected in breast cancer. Based on previous proteomic and transcriptomics analysis, carboxypeptidase B1 was identified as a potential biomarker of lymph node metastasis in luminal A breast cancer. The first aim of this thesis was to analyze the impact of CPB1 expression on cancer cell properties, primarly stiffness, that had been analysed by atomic force microscopy. It was found that an increased level of CPB1 causes a significant decrease of MCF-7 cancer cell stiffness, similarly, these cells were morphologically differrent from the cells that do not express CPB1 protein. Further in the thesis, the effect of a panel of potential antimetastatic inhibitors on CPB1 level was tested, in order to characterise the mechanism of CBP1 prometastatic effect and to outline possible ways of therapeutical modulation. The level of CPB1 was significantly affected by two inhibitors. Norcantharidin significantly decreased CBP1 level after 24 hours. On the other hand, ATM- protein inhibitor KU 60019 increased CPB1 level after 24 hours. Both inhibitors are supposed to lead cells to apoptosis, which was tested by flow cytometry. While norcantharidin led MCF-7 cells to apoptosis and late apoptosis, KU 60019 caused necrosis of the MCF-7 CPB1 cells, but not the apoptosis. Furthermore, the MCF-7 CPB1 cells proteome was compared with that of the control MCF-7 GFP line cells using LC-MS/MS in data independent acquisition (DIA) mode. In the MCF-7 CPB1 line, there was an increased level of epithelial-mesenchymal transition markers, increased levels of proteins stimulating cancer cell aggression and angiogenesis and decreased levels of proteins responsible for cell adhesion. This data indicate a possible relationship of CPB1 to epithelial-to-mesenchymal transition and indicate possible means of modulation of its protein level.
