Genes of sulfur metabolism in the biooxidation of pyrite

Abstract:

Tato diplomová práce se zabývá identifikací genů kódujících proteiny, které se účastní oxidace elementární síry a redukovaných anorganických sirných sloučenin u bakterie Acidithiobacillus ferrooxidans a jejich exprese při oxidaci pyritu. V teoretické části jsou shrnuty současné poznatky o buněčné stavbě bakterie At. ferrooxidans a předpokládané metabolické dráhy oxidace železa(II) a zejména sirných látek. Dále se tato část zaměřuje na metody molekulární analýzy na úrovni transkripce. V experimentální části byla optimalizována kultivace bakterií At. ferrooxidans na pyritu a následná separace planktonických a adherovaných buněk. Byly purifikovány proteiny tetrathionanhydrolasa a thiosírandehydrogenasa účastnící se metabolismu sirných látek, u nichž byly identifikovány jejich geny. Dále byla sledována exprese těchto a dalších vybraných genů sirného metabolismu v kulturách At. ferrooxidans rostoucích na pyritu.

Abstract:

This thesis is focused on the identification of genes encoding proteins that are involved in the oxidation of elemental sulfur and reduced inorganic sulfur compounds and their expression in Acidithiobacillus ferrooxidans grown on pyrite. The theoretical part summarizes the current knowledge about cellular structure of At. ferrooxidans and proposed pathways of iron oxidation and especially oxidation of sulfur compounds. Furthermore, this section focuses on methods of molecular analysis used at a transcription level. In the experimental part were optimized cultivation of At. ferrooxidans on pyrite and separation of planktonic and sessile cells. The proteins tetrathionate hydrolase and thiosulfate dehydrogenase involved in the metabolism of sulfur compounds were purified and their genes have been identified. The expression profiles of identified genes and some other genes supposed to be involved in sulfur metabolism in At. ferrooxidans grown on pyrite were examined.
