Isolation of Cellulose-Degrading Bacteria and Determination of Their Cellulolytic Potential

Abstract:

Celulolytické bakterie patří mezi nedílné součásti globálního cyklu uhlíku. Spolu s ostatními celulolytickými organismy degradují velmi odolnou celulózu na jednoduché sacharidy. Toho dosahují prostřednictvím specifických, jimi produkovaných enzymů. Celulolytické bakterie samozřejmě nenachází využití pouze v klíčových přírodních procesech, ale také v průmyslu. V této práci byl proveden screening a stanovení celulolytické aktivity psychrofilních a mezofilních celulolytických bakterií izolovaných ze stojatých povrchových vod. Screening a stanovení celulolytické aktivity probíhalo na selektivním karboxymethyl celulózového (CMC) agaru v kombinaci se zobrazením projasněných zón Lugolovým roztokem. Z celkového počtu 90 izolátů (50 mezofilních a 40 psychrofilních) bylo 40 % mezofilních a 38 % psychrofilních izolátů celuloticky aktivních. U celulolytických izolátu byla následně stanovena kvantitativní hodnota celulolytické aktivity vyjádřená jako enzymatický index. Enzymatický index se u mezofilních izolátů pohyboval v rozmezí 0,31 až 3,72 a u psychrofilních izolátů v rozmezí 0,46 až 6,23. Celulolytické izoláty byly nakonec charkterizovány pomocí Gramova barvení a KOH testu, přičemž jak u mezofilních, tak u psychrofilních celulolytických izolátů převažovaly gramnegativní tyčinky.

Abstract:

Cellulolytic bacteria belong to integral components of the global carbon cycle. They degrade very resistant cellulose into simple carbohydrates together with other cellulotyc organisms. This process is facilitated by specialized enzymes that they produce. Except their ecological importance, these bacteria have also industrial utilization potential. This study focused of screening and assessment of cellulolytic activity in psychrophilic and mesophilic cellulolytic bacteria isolated from stagnant waters. The evaluation was conducted using selective carboxymethylcellulose (CMC) agar, complemented by Lugol's solution to visualize the zones of clearance. Out of 90 isolates (50 mesophilic and 40 psychrophilic), 40% of the mesophilic and 38% of the psychrophilic bacteria exhibited cellulolytically active. The enzymatic index, reflecting the extent of cellulolytic activity, was measured for these active isolates. It varied from 0.31 to 3.72 among mesophilic isolates and from 0.46 to 6.23 for psychrophilic isolates. The cellulolytic bacteria were further characterized using by Gram staining and the KOH test, demonstrating a predominance of Gram-negative rods in both temperature groups.
