Study of plant telomerase components

Abstract:

Telomeráza je ribonukleoproteinový komplex podílející se na udržování integrity telomer, nukleoproteinových struktur na koncích eukaryotických chromozomů. Telomery chrání konce chromozomů před ztrátou genetické informace po mnohonásobné replikaci a odlišují je od dvouvláknových zlomů na DNA. Telomeráza se skládá ze dvou hlavních podjednotek – telomerázové reverzní transkriptázy (TERT) a telomerázové RNA (TR). Telomerázová RNA poskytuje RNA templát pro syntézu telomerických DNA repetic. Znalost sekvence a sekundární struktury telomerázové RNA u rostlin je velmi recentní poznatek. V roce 2019 byly identifikovány TR u rostlin, byl zjištěn jejich monofyletický původ a jejich biogeneze odlišná od do té doby známých TR obratlovců a kvasinek. Ukazuje se, že nejen délka a nukleotidové složení, ale také prostorové uspořádání TR ovlivňuje vazbu proteinů a umožňuje sestavení funkčního komplexu telomerázy. V rámci diplomové práce byla optimalizována metoda ChIRP-MS (Comprehensive identification of RNA-binding proteins by mass spectrometry), patřící mezi RNA-centric metody zkoumání protein-RNA interakcí, pro použití v rostlinných systémech. Následně byla využita pro identifikaci proteinů potenciálně interagujících s TR a byly tak získány kandidátní proteiny pro další zkoumání pomocí jiných metod a studium biogeneze TR.

Abstract:

Telomerase is a ribonucleoprotein complex which is responsible for maintaining the integrity of telomeres, nucleoprotein structures at the ends of eukaryotic chromosomes. Telomeres protect the ends of chromosomes against the loss of genetic information after repeated replication and make them distinguishable from DNA double-strand breaks. Telomerase comprises two main subunits – telomerase reverse transcriptase (TERT) and telomerase RNA (TR). Telomerase RNA provides an RNA template for the synthesis of telomeric DNA repeats. The knowledge of the sequence and the secondary structure of telomerase RNA in plants is a very recent piece of information. In 2019, plant TRs were identified and it was found that their origin is monophyletic and the biogenesis differs from then known vertebrate and yeast TRs. It turns out that not only length and nucleotide composition but also the space organization of TR has an impact on protein binding and enables the assembly of a functional telomerase complex. In this diploma thesis, ChIRP-MS (Comprehensive identification of RNA-binding proteins by mass spectrometry), an RNA-centric method for studying protein-RNA interactions, was optimized for plant systems. The method was used for the identification of potential interacting proteins of TR which can help to elucidate telomerase complex composition and biogenesis.
