Interactions of telomeric proteins and transcription factors

Abstract:

U převážné většiny eukaryotických organismů je DNA uložena v jádře ve formě lineárních chromozomů. Koncové části chromozomů se nazývají telomery. Telomery jsou nukleoproteinové komplexy, které mají za úkol chránit konce chromozomů před nukleolytickou degradací, opravnými mechanismy buňky a nechtěnou fúzí chromozomů. Po každé replikaci dochází ke zkracování telomer, které může vést až k buněčnému stárnutí a následné apoptóze. Tento proces však umí zvrátit enzym telomeráza, který je schopen připojovat nukleotidy ke koncům telomerové DNA. Na regulaci aktivity telomerázy se podílí komplex proteinů nazývaný shelterin. Shelterin vytváří na telomerách lasovité struktury, které ovlivňují přístup telomerázy a dalších enzymů. Takto shelterin reguluje délku telomer a podílí se na procesech stárnutí a nádorové transformace buněk. K nádorové transformaci buněk může také přispívat transkripční faktor REST. REST je represorem exprese více než stovky genů a reguluje tak diferenciaci zejména nervových buněk, zajišťuje jejich sebeobnovu a udržuje je v dormálním stavu. Při nekontrolovaném vývoji se REST může změnit v onkogen. Tato práce se zabývá zkoumáním interakčních oblastí centrálního proteinu shelterinu TRF2 a transkripčního faktoru REST pomocí měření změn intenzity fluorescence.

Abstract:

For the majority of eukaryotic organisms, DNA is stored in the nucleus in the form of linear chromosomes. Chromosome ends are called telomeres. Telomeres are nucleoprotein complexes that protect chromosome ends from nucleolytic degradation, repair processes of the cell and unwanted fusion of chromosomes. After each replication telomeres reduce their length which can lead to cell aging and subsequent apoptosis. Enzyme called telomerase can reverse telomere shortening. Telomerase is able to add nucleotides to the ends of telomeric DNA. Protein complex called shelterin participate in the regulation of telomerase activity. Shelterin forms lariat structures on telomeres which prevent access of telomerase and other enzymes to telomeric DNA. Thus shelterin regulates telomere length and participates in processes of cell aging and tumor transformation. Similarly, transcription factor REST can also contribute to tumor cell transformations. REST is a repressor of expression of more than one hundred of genes. REST regulates differentiation of especially neural cells, it provides self-renewal capacity and keeps cells in dormant state. During uncontrolled development, REST can turn to an oncogene. This work is focused on research of interaction parts of telomeric protein TRF2 and transcriptional factor REST by measuring the fluorescence intensity changes.
