Assessment of the Influence of Antimicrobial Agents on the Microbial Biofilm by Fluorescent Spectrometry

Abstract:

Biofilm je společenstvo mikroorganismů uchycené k určitému povrchu, uložené v extracelulární matrix, což je hmota, kterou si toto společenstvo samo aktivně vytváří. Podle zprávy National Institutes of Health z roku 2007 je až 80 % infekcí člověka spojeno s biofilmy. Mezi nejzávažnější patří zejména infekce krevního řečiště spojené s přítomností katetrů, kanyl a cévních či kloubních náhrad. Dále působí těžké pneumonie, subakutní endokarditidy, osteomyelitidy nebo otitidy. Naopak nejčastějším onemocněním, které je spojeno s přítomností biofilmu zůstává zubní kaz. Tato diplomová práce bude posuzovat vliv různých látek antimikrobiální povahy (antibiotik, fágových preparátů apod.) na biofilmy klinicky významných druhů mikrobů. Hodnocení vlivu bude probíhat jak standardními metodami, tak zejména s využitím fluorescenční spektrometrie, jako relativně nové metody hodnocení. Výsledky poslouží jako zdroj informací o účinnosti různých antimikrobiálních látek na mikrobiální biofilmy. Dále práce objasní současné možnosti využití fluorescenční spektrometrie jako alternativní metody ke standardním postupům v klinické praxi. Po formální stránce bude práce upravena dle Instrukcí pro vypracování diplomové práce.

Abstract:

Biofilm is a community of microorganisms attached to a particular surface deposited in an extracellular matrix, which is a mass that this community actively creates. According to the 2007 National Institutes of Health report, up to 80% of human infections are associated with biofilms. Bacterial infections associated with the presence of catheters, cannula, and vascular or articular replacements are among the most serious. It also causes severe pneumonia, subacute endocarditis, osteomyelitis or otitis. On the contrary, the most common disease associated with the presence of biofilm remains tooth decay. This diploma thesis will examine the influence of various antimicrobial substances (antibiotics, phage preparations, etc.) on biofilms of clinically important microbe species. Impact assessment will be conducted both using standard methods and using fluorescence spectrometry as relatively new assessment methods. The results serve as a source of information on the efficacy of various antimicrobials on microbial biofilms. In addition, the thesis explains the current possibilities of using fluorescence spectrometry as an alternative method to standard procedures in clinical practice. Formally, the work will be modified according to the Instruction for the Diploma Thesis.
