Molecular and biochemical characterization and localization of peptidase inhibitors from Eudiplozoon nipponicum

Abstract:

Eudiplozoon nipponicum (Rodina Diplozoidae, Polyophisthocotylea) je hematofágní ektoparazit žijící na žábrách kapra obecného (Cyprinus carpio). Zástupci třídy Monogenea se obvykle živí slizem či epiteliálními buňkami kůže či žaber. Hematofágie je v tomto ohledu spíše výjimečnou strategií. Ačkoliv ztráta krve většinou není pro rybu fatální, poškozené lamely žaber často představují vstupní bránu pro případné sekundární mikrobiální infekce, které mohou způsobit snížení fitness ryb a následně finanční ztráty v chovech. V rámci interakce parazit-hostitel by výzkum molekul klíčových pro přežití parazita (např. zapojených do příjmu potravy, úniku před imunitním systémem apod.) mohl umožnit lépe porozumět těmto molekulárním interakcím, a to nejen v případě monogeneí, ale i u jiných skupin parazitů. Navzdory zřejmému potenciálu, vlastnosti funkčních proteinů, např. peptidáz a jejich inhibitorů jsou v rámci platyhelmintů u zástupců monogeneí nejméně probádané. Tato práce sumarizuje informace dostupné v literatuře zabývající se inhibitory peptidáz platyhelmintů, které jsou prezentovány na několika zástupcích trematod, cestod a monogeneí. První část práce je zaměřena na tři důležité skupiny inhibitorů: serpiny, cystatiny a Kunitz inhibitory platyhelmintů, které byly identifikovány a následně charakterizovány také u zástupce třídy Monogenea – E. nipponicum. Do této oblasti spadá první ze dvou prvoautorských publikací, na kterých je tato práce postavena. V této studii byl poprvé charakterizován inhibitor serinových peptidáz (serpin) E. nipponicum (EnSerp1). Byly popsány jeho sekvenční a strukturní vlastnosti a také inhibiční potenciál proti několika vybraným serinovým peptidázami (trypsin, faktor Xa, kalikrein a plazmin). Dále byl EnSerp1 identifikován v exkrečně-sekrečních produktech E. nipponicum, což poukazuje na jeho pravděpodobné zapojení do procesů krmení. V další studii související s disertační prací (spoluautorská práce), byl zkoumán inhibitor cysteinových peptidáz (cystatin) E. nipponicum. Kromě hlavního inhibičního místa pro papainové peptidázy, analýza aminokyselinové sekvence odhalila i přítomnost druhého neobvyklého inhibičního místa pro asparaginylovou endopeptidázu. Navíc byl prokázán inhibiční účinek cystatinu na trávení hemoglobinu in vitro. Druhá část práce se věnuje technikám umožňujícím lokalizaci biomolekul, především proteinů (peptidáz, jejich inhibitorů), ve tkáních organismu. V této části jsou definovány čtyři nejčastěji využívané přístupy – imunohistochemie, in situ hybridizace, zobrazovací hmotnostní spektrometrie a laserová mikrodisekce kombinovaná s analýzou pomocí kapalinové chromatografie s tandemovou hmotnostní spektrometrií (LCM-LC-MS/MS). Možnosti a limitace každé techniky jsou diskutovány s ohledem na využití v parazitologii. Do této oblasti spadá druhá prvoautorská publikace, která sumarizuje výsledky dosažené s využitím LCM-LC-MS/MS. Z kryořezů celých dospělců E. nipponicum, byly pomocí mikrodisekce (LCM) dále izolovány vzorky tří tkání (střevo, tegument a parenchym), které byly následně analyzovány (LC-MS/MS). Tímto způsobem byly připraveny tkáňově specifické proteomy, přičemž výlučně ve střevě byla potvrzena přítomnost několika dříve popsaných katepsinů E. nipponicum. Získaná data navíc představují první rozsáhlejší proteomický dataset nejen pro E. nipponicum, ale rovněž pro celou třídu Monogenea.

Abstract:

Eudiplozoon nipponicum (family Diplozoidae, Polyopisthocotylea) is hematophagous ectoparasite from the gills of common carp (Cyprinus carpio). For members of class Monogenea is common feeding on mucus or epithelial cells of skin or gills. Hematophagy is much more invasive and rather rare strategy in this respect. While the blood loss is not usually fatal for the fish, damaged gill lamellae represent entry point for secondary microbial infections, which may result in reduction of fish fitness and subsequently financial losses in aquacultures. Therefore, research of this parasite-host interaction related to molecules which are employed and might be crucial for parasite survival (e.g. feeding, evasion to immune system etc.), would broaden our understanding of such molecular communication in case of monogeneans and also other groups of parasites. Despite the evident potential, the properties of functional proteins, e.g. peptidases and inhibitors of monogeneans, are among the less investigated in whole phylum Platyhelminthes. This thesis summarizes available literature dealing with peptidase inhibitors of platyhelminths presented on several selected members of trematodes, cestodes and monogeneans. In the first part of the thesis, the attention is payed to three important inhibitor groups: serpins, cystatins and Kunitz-type inhibitors of platyhelminths, which has been identified and also characterized for member of class Monogenea E. nipponicum. To this research area fits the first of two original publications on which the thesis is build. In this study the first serine peptidase inhibitor (serpin) of E. nipponicum named EnSerp1 was characterized. Its sequential and structural properties were examined as well as the inhibitory potential against several serine peptidases (trypsin, factor Xa, kallikrein, and plasmin). It was further identified in excretory-secretory products of E. nipponicum suggesting the probable role in feeding. In other study related to the thesis (co-authored work), cysteine peptidase inhibitor (cystatin) of E. nipponicum was examined. Analysis of sequence revealed presence of unusual inhibitory site for asparaginyl endopeptidase, apart from the “main” one targeted on papain-like peptidases. Moreover, cystatin was proved to have inhibitory effect on hemoglobin digestion in vitro. Second part of the thesis is dedicated to techniques used for localization of various biomolecules, predominantly proteins (peptidases, inhibitors), in tissue sections. Four general techniques were described, immunohistochemistry, in situ hybridization, mass spectrometry imaging and laser microdissection combined with liquid chromatography tandem mass spectrometry (LCM-LC-MS/MS). Possibilities and limitations of each technique are discussed and confronted with the records of its utilization in parasitology. To this area, the second original publication is aimed. It describes the latter method applied for dissection of three types of tissue (gut, tegument and parenchyma) from E. nipponicum cryo-sections with subsequent LC-MS analysis. Through generation of tissues specific proteomes, also the location of several already described E. nipponicum cathepsins (L and B) have been exclusively confirmed in gut. Moreover, obtained proteomic data in this study does not only show the location of identified proteins, but forms the first comprehensive proteomic dataset available for the whole class of Monogenea.
