Isolation and characterization of staphylococcal polyvalent bacteriophage mutants

Abstract:

Bakterie rodu Staphylococcus jsou běžnou součástí mikroflóry člověka i zvířat, avšak v důsledku oslabení imunitního systému hostitele mohou být původci závažných infekcí. Vzhledem ke globálnímu rozšíření kmenů rezistentních k mnoha dostupným antibiotikům se nabízí možnost léčit tyto infekce pomocí virulentních bakteriofágů. Pro fágovou terapii je vhodné využití polyvalentních bakteriofágů, které jsou schopné napadat velké množství kmenů. V této práci byly izolovány tři mutantní stafylokokové bakteriofágy s rozšířeným spektrem hostitelských kmenů, jejichž genomy se odlišují od genomů původních fágů přítomností několika delecí a inzercí. Analýzou kmenů, na kterých byli noví mutanti izolováni, bylo zjištěno, že součástí genomů všech tří těchto kmenů jsou profágy a plazmidy, což je u pomnožovacích kmenů terapeutických fágů nežádoucí. Vzhledem k rozšířenému množství kmenů stafylokoků, které jsou nové mutantní fágy schopny lyzovat, je jejich budoucí využití v terapii stafylokokových infekcí výhodné. Pro jejich pomnožování však bude nutno nalézt jiný, bezpečný kmen se známým genomem prostým profágů i plazmidů, aby tak byla zajištěna zdravotní nezávadnost fágového preparátu.

Abstract:

The bacteria of the genus Staphylococcus are common part of human and animal microflora, but when the immune system of the host is weakened, they can become the agents of serious infections. Due to global distribution of strains resistant to several antibiotics, there is a possibility to cure these infections by virulent bacteriophages. For the phage therapy it is more efficient to use polyvalent bacteriophages able to infect large number of strains. In this work three mutant staphylococcal bacteriophages with extended spectrum of host strains were isolated, whose genomes differ from genomes of the original phages by presence of several deletions and insertions. By analysis of the strains from which the new mutants were isolated, it was found that prophages and plasmids are parts of the genomes of all three these strains. Presence of such genetic elements in the strains is unwanted and may hamper their use as therapeutic agents. Thanks to the extended number of strains of staphylococci the new mutants are able to lyse, they seem to be suitable for potential use in the therapy of staphylococcal infections. To provide harmlessness of the phage preparation it will be, however, necessary to find another safe strain with known genome sequence lacking prophages and plasmids for the purpose of propagation of the phages.
