The significance of hypoxia in cancer microenvironment

Abstract:

This thesis is focused on the impact of hypoxia in a tumor microenvironment. The first part of the thesis is focused on the introduction to the problematics, basic definitions of hypoxia and its detection possibilities. In the second part, there are described different types of metabolic changes in cancer cells, which arise under hypoxic stress conditions. Metabolic pathways both of cancer cells and normal tissue cells are compared in this part. The third part is devoted to the most important genetic and proteomic changes occuring in the tumor cells during hypoxia. The major attention is paid to the changes in gene expression and to the impact of these changes on particular cells. In the last part, some options of hypoxic tumor regions reduction are mentioned, because the oxygen deficit in the tumor tissue is associated with increased agresivity of the disease and worse prognosis for patients.

Abstract:

Táto práca sa venuje problematike vplyvu hypoxie na nádorové bunky a ich mikroprostredie. Prvá časť je venovaná úvodu do problematiky, základným charakteristikám hypoxie ako aj možnostiam jej detekcie v tkanivách. V nadväzujúcej druhej časti sú popísané rôzne druhy metabolických zmien nádorových buniek, ktoré vznikajú pod vplyvom stresujúcich podmienok nedostatku kyslíka. Metabolické procesy nádorových buniek sú porovnávané s procesmi v zdravých bunkách človeka. Tretia časť je venovaná najvýznamnejším genetickým a proteomickým zmenám prebiehajúcim v nádorových bunkách počas hypoxie. Pozornosť je venovaná hlavne zmene expresie jednotlivých génov a následnému dopadu týchto zmien na mikroprostredie buniek. V poslednej časti sú spomenuté možnosti redukcie hypoxických oblastí v nádoroch, keďže nedostatok kyslíka v tkanive je spojený so zvýšenou agresivitou ochorenia a so zhoršenou prognózou pacientov.
