Mutual interactions of telomerase-associated proteins

Abstract:

The aim of this bachelor thesis was to contribute to a more detailed elucidation of the structure of the RNA subunit of the telomerase enzyme (AtTR) in plant cells which was described recently in our laboratory. Homologues of the p65 protein belonging to the LA family of proteins derived from Tetrahymena thermophila and the human nucleolin protein, were selected as possible candidates associated with AtTR in the model organism Arabidopis thaliana. Homologues of LA proteins have been described in A. thaliana cells, as has the nucleolin protein, but their association with AtTR has not yet been a subject of research. From an evolutionary point of view, LA proteins are of particular interest, as plants and ciliates are more evolutionarily related than, for example, plants with mammals or yeast. The theoretical part of the work briefly summarizes the current knowledge about telomeres, telomerase, and its differences in structure across human, plant, yeast, and ciliate cells. The practical part is focused on the cloning of selected protein homologues into appropriate destination vectors, thanks to which they will be used in the future to study the structure of the RNA subunit in A. thaliana.

Abstract:

Cieľom tejto bakalárskej práce bolo prispieť k detailnejšej charakterizácii štruktúry RNA podjednotky enzýmu telomerázy (AtTR) v rastlinných bunkách, ktorá bola popísaná v našom laboratóriu len nedávno. Homológy proteínu p65 patriaceho do rodiny LA proteínov, pochádzajúceho z Tetrahymena thermophila a ľudského proteínu nukleolínu, boli na základe analýzy vybratí ako možní kandidáti asociovaní s AtTR u modelového organizmu Arabidopis thaliana. Homológy LA proteínov boli v bunkách A. thaliana popísané, tak isto ako aj proteín nukleolín, ale ich asociácia s AtTR ešte nebola doposiaľ predmetom výskumu. Z evolučného hľadiska sú zaujímavé najmä LA proteíny, keďže rastliny a nálevníci sú evolučne príbuznejší než napríklad rastliny s cicavcami, či kvasinkami. Teoretická časť práce stručne zhrňuje súčasné poznatky o telomérach, telomeráze a jej rozdieloch v štruktúre naprieč bunkami ľudskými, rastlinnými, kvasiniek a nálevníkov. Praktická časť je zameraná na klonovanie vybraných homológov proteínov do príslušných destinačných vektorov, vďaka čomu budú v budúcnosti použité pri skúmaní štruktúry RNA podjednotky u A. thaliana.
