Optimization of cancer vaccines based on dendritic cells

Abstract:

Dendritické buňky patří mezi antigen-prezentující buňky imunitního systému. Jejich funkcí je pohlcovat a zpracovávat antigeny ze svého okolí, vystavovat je na svém povrchu a takto je zpřístupňovat dalším buňkám imunitního systému, zejména T-lymfocytům. Ty pak zahajují specifickou reakci vůči buňkám a částicím nesoucím prezentované antigeny. Charakter imunitní odpovědi se odvíjí od dalších signálů, vysílaných dendritickými buňkami T-lymfocytům v průběhu prezentace antigenů. Signály vysílané dendritickými buňkami se liší v závislosti na složkách prostředí, v němž dendritické buňky dozrávaly (maturovaly). Předkládaná dizertační práce je zaměřena na charakterizaci dendritických buněk připravených diferenciací krevních monocytů a maturovaných různými způsoby. Cílem práce je optimalizovat přípravu dendritických buněk pro protinádorovou vakcinaci a vybrat maturační strategii, stimulující nejvhodnější vlastnosti pro protinádorovou imunoterapii. Dílčími cíli práce je zejména srovnání vlivu různých kombinací maturačních cytokinů na důležité imunostimulační vlastnosti dendritických buněk - zralost (reprezentovanou expresí specifických povrchových molekul), produkci cytokinů, migraci dendritických buněk, schopnost aktivace T-lymfocytů a efektivitu aktivovaných T-lymfocytů v cytotoxické reakci s cílovými nádorovými buňkami. Ze šesti testovaných maturačních strategií jsme jako optimální pro protinádorovou imunoterapii identifikovali maturaci pomocí interferonu-γ a lipopolysacharidu. Tento způsob maturace produkuje plně zralé dendritické buňky, stimulující autologní i alogenní T-lymfocyty k nejvyšší proliferaci i produkci cytokinů, a zároveň indukující cytotoxickou reakci vůči cílovým nádorovým buňkám, nikoliv však vůči kontrolním buněčným liniím, exprimujícím odlišné antigeny. Použití takto maturovaných dendritických buněk v protinádorové imunoterapii by tedy mohlo mít značný přínos pro léčbu pacientů s různými typy nádorových onemocnění.

Abstract:

Dendritic cells belong to antigen-presenting cells of the immune system. Their function is to absorb and process antigens from their surroundings, display them on their surface and thus to make them accessible for other cells of the immune system, mainly T-cells. T-cells subsequently induce specific reaction to the cells or particles carrying presented antigens. Character of the immune response is dependent on additional signals, provided by dendritic cells to T-cells during the process of antigen presentation. Signals expressed by dendritic cells differ according to composition of the environment, which was present during their maturation. Submitted dissertation thesis is focused on characterization of dendritic cells, prepared by differentiation of blood monocytes, and matured by different maturation strategies. The aim of this work was optimization of dendritic cell preparation for cancer immunotherapy and identification of maturation strategy generating dendritic cells with optimal characteristics for cancer immunotherapy. The work compares the influence of different combinations of maturation cytokines on important immunostimulatory characteristics of dendritic cells - mature state (represented by expression of specific surface molecules), cytokine production, migratory capacity of dendritic cells, their capability of T-cell stimulation and the efficiency of stimulated T-cells in a cytotoxic reaction against specific tumour cells. Among the six maturation cocktails tested, we identified the combination of interferon-γ and lipopolysaccharide as the most appropriate for cancer immunotherapy. This maturation strategy generates fully mature dendritic cells, which stimulate both autologous and allogeneic T-cells and subsequently induce cytotoxic reaction against target tumour cells, but not control cell lines expressing different antigens. Use of this maturation cocktail for tumour immunotherapy could bring significant benefits for treatment of patients with various types of cancer.
