Factors affecting the regulation of ferA gene expression in the bacterium Paracoccus denitrificans

Abstract:

K detekci genové exprese ferA v bakterii Paracoccus denitrificans byl využit reportérový enzym β-galaktosidasa. Experimenty byly prováděny v aerobním médiu v prostředí s nízkou koncentrací železa a v přítomnosti Fe(III)-(DHB)3. Postupná limitace železa pro bakterii znamenala zpomalení jejího růstu. Obdobné vlastnosti vykazoval kmen s přerušeným ferA. Vypočtené aktivity β-galaktosidasy v závislosti na čase a vzrůstajícím zákalu bakteriální kultury postupně klesaly. Pro nejnižší koncentraci železa v prostředí se aktivita reportérového enzymu neměnila. Kmen s přerušeným ferA v přítomnosti Fe(III)-(DHB)3 vykazoval téměř identický růst jako v podmínkách s limitujícími koncentracemi železa. Aktivita β-galaktosidasy pro nejvyšší koncentraci Fe(III)-(DHB)3 se neměnila jako v případě nejnižší koncentrace volného železa. Exprese genu dosahuje relativně nejvyšších hodnot s dostatečnou limitací železa nebo při nejvyšší použité koncentraci Fe(III)-(DHB)3.

Abstract:

For the detection of gene expression ferA in bacteria Paracoccus denitrificans reporter enzyme β-galactosidase was used. Experiments were performed in an aerobic medium in an environment with low concentration of iron and in the presence of Fe(III)-(DHB)3. Gradual limitation of iron caused reduction of the growth of bacteria. Similar behaviour was observed at strain with interrupted ferA. Depending on the time and increasing turbidity of bacterial cultures, calculated β-galactosidase activity was decreased gradually. Activity of reporter enzyme has not changed for the lowest concentration of iron in the environment. Strain with interrupted ferA in the presence of Fe(III)-(DHB)3 showed almost identical growth conditions like in the condition with limited concentrations of iron. β-galactosidase activity for the highest concentration of Fe(III)-(DHB)3 has not changed as in the case of low concentrations of the free iron. Gene expression reaches relative the highest values with sufficient iron limitation or with the usage of the highest concentration of Fe(III)-(DHB)3.
