Abstract:

Tato diplomová práce se věnuje genetickému pozadí zubního kazu u dočasné-ho a stálého chrupu. Zubní kaz je velmi rozšířená multifaktoriální choroba, jejíž vznik a rozvoj je ovlivněn přítomností kariogenních bakterií v zubním plaku, velkým množstvím fermentovatelných sacharidů v potravě či ústní hygienou. Náchylnost k zubnímu kazu může být geneticky podmíněna, zkoumány jsou mj. geny kódující proteiny spojené s morfologií zubní tkáně. Na základě literární rešerše byly vybrány jednonukleotidové polymorfizmy (SNPs) v genech kódujících akvaporiny (AQPs), což jsou selektivní vodní kanály, které svou funkcí mohou ovlivnit správný vývoj zubu i sekreci slin. V této práci byly zavedeny metodiky pro stanovení vybraných SNPs v AQPs a dále byly provedeny genetické asociační studie u české populace i v návaznosti na Bakalářskou práci, která byla věnována optimalizaci metodik pro stanovení SNPs v genech pro další proteiny zapojené při tvorbě tvrdých zubních tkání. SNPs v genech pro amelogenin (AMELX), kalikrein 4 (KLK4) a v AQPs byly asociovány se zubním kazem u dočasného a/nebo stálého chrupu u českých dětí a tato zjištění přispívají k objasnění etiopatogeneze zubního kazu.

Abstract:

This diploma thesis deals with the genetic background of dental caries in primary and permanent dentition. Tooth decay is a widespread multifactorial disease, which origin and development is affected by the presence of cariogenic bacteria in dental plaque, large amounts of fermentable carbohydrates in the diet or oral hygiene. The susceptibility to dental caries may be genetically determined, among others, genes encoding proteins associated with dental tissue morphology are investigat-ed. Single nucleotide polymorphisms (SNPs) in genes encoding aquaporins (AQPs) were selected based on the literature search. AQPs are selective water channels that can affect the tooth development and the saliva secretion. In this thesis, the methodologies for the determination of selected SNPs in AQPs were designed and genetic association studies were performed in the Czech population in continuity with the Bachelor thesis, which was devoted to the optimalization of methodologies for determination of SNPs in genes encoding proteins involved in the formation of hard dental tissues. SNPs in genes for amelogenin (AMELX), kallikrein 4 (KLK4) and in AQPs were associated with dental caries in primary and/or permanent dentition in Czech children and these findings contribute to the knowledge of the etiopathogenesis of dental caries.
