Possibilities of isolation and identification Amanita muscaria and Amanita pantherina Toxins by LC/MS

Abstract:

Předkládaná disertační práce se zabývá izolací a průkazem toxinů muchomůrky červené a tygrované v biologickém materiálu (moč, sérum, žaludeční obsah). V posledních letech se do popředí zájmu především mladých toxikomanů dostává experimentování s psychotropními druhy hub. Mezi tyto houby řadíme i muchomůrku červenou a tygrovanou (Amanita muscaria a Amanita pantherina). Zatímco u muchomůrky červené jde nejčastěji o úmyslnou konzumaci s cílem navození halucinogenního stavu, u muchomůrky tygrované se jedná většinou o náhodnou intoxikaci, způsobenou její záměnou s jedlými druhy hub. Hlavními obsahovými látkami těchto hub jsou ibotenová kyselina a muscimol (deriváty isoxazolu) a muskarin. Tyto látky, velmi podobné neurotransmiterům v mozku, jsou schopny vyvolat neurotoxické symptomy u uživatelů. Přestože intoxikace těmito houbami jsou zřídka smrtelné, je důležité jejich včasné rozpoznání a zahájení léčby. Problémem v soudobé klinické a forensní toxikologii je absence objektivní analytické metody pro průkaz a stanovení účinných látek těchto hub z biologického materiálu intoxikovaných. Doposud se k diagnostice otrav používají subjektivní mikroskopické metody. Cílem bylo vytvořit objektivní LC-MS metodu pro rychlou diagnostiku intoxikace těmito houbami. Experimenty byly provedeny na přístroji LC-MS-ESI. V metodě pro průkaz toxinů byla měření provedena na koloně Gemini C18 s užitím 8 mmol.l-1 heptafluoromáselné kyseliny jako mobilní fáze. Za těchto chromatografických podmínek byly sledovány ionty m/z 159 pro ibotenovou kyselinu, m/z 115 pro muscimol a m/z 174 pro muskarin. V metodě pro stanovení muskarinu byla měření provedena na koloně Gemini C18 s užitím mobilní fáze složená z 8 mmol.l-1 heptafluoromáselné kyseliny a acetonitrilu. Sledované ionty byly m/z 174 pro muskarin a m/z 216 pro acetylovaný muskarin. Pro izolaci toxinů z moče byla použita extrakce na pevné fázi, kolonka Strata X-CW pro muskarin a Discovery SCX pro ibotenovou kysolaci a muscimol. Podařilo se nalézt citlivou a specifickou metodu kapalinové chromatografie s hmotnostní detekcí pro analýzu hlavních toxinů muchomůrek červené a tygrované - ibotenovou kyselinu, muscimol a muskarin. Metodu lze rovněž použít při objasňování intoxikací způsobených jinými druhy hub, obsahujících tyto toxiny.

Abstract:

The present thesis is focused on isolation and identification Amanita muscaria and Amanita pantherina toxins in biological material, such as urine, blood or gastric content. Over the past years, experimenting with psychotropic mushrooms, especially among young drug addicts, has been brought to the foreground of interest. Among such mushrooms we include Amanita muscaria and Amanita pantherina as well. While Amanita muscaria is deliberately consumed with the aim of evoking a hallucinogenic state, with Amanita pantherina it is mostly an accidental intoxication, when mistaken for some edible mushrooms. The major toxins contained in these Amanitas are ibotenic acid and muscimol (isoxazole derivates) and muscarine. These toxins, very similar to nerve transmitters in the brain, are able to give cause for hallucinogenic symptoms in their users. Although intoxications through these mushrooms are rarely lethal, it is important to determine them soon and initiate a medical treatment. The problem of today`s forensic toxicology is the absence of an objective analytical method for identification and determination of the mushrooms` toxins from biological materials of patients. Nowadays, the diagnosis of these mushroom poisonings is almost entirery determined by subjective microscopic examinations. The aim of the project was to elaborate and introduce an objective LC-MS method for rapid and reliable diagnosis of intoxications by these mushroom toxins. The experiments were carried out using an LC-MS equipped with electrospray ionization. In the method for toxins identification, the separation of the toxins was achieved on a Gemini C18 column with 8 mmol.l-1 heptafluorobutyric acid as the mobile phase. Under the described chromatographic conditions, intensive ions at m/z 159 for ibotenic acid, m/z 115 for muscimol and m/z 174 for muscarine were observed. In the method for muscarin determination, the measurement was achieved on a Gemini C18 column with 8 mmol.l-1 heptafluorobutyric acid and acetonitril as the mobile phase. Under the described chromatographic conditions, intensive ions at m/z 174 for muscarine and m/z 216 for acetyled muscarine were observed. Solid phase extraction was applied for isolation of the toxins from urine, Strata X-CW column for muscarine and Discovery SCX column for ibotenic acid and muscimol. A sensitive and specific liquid chromatography-mass spectrometry assay was developed for analysis of principal toxins of A. muscaria and A. pantherina. The method can be used for explication of intoxication caused by another mushrooms containes the same toxins.
