A novel post-translational modification of Dishevelled

Abstract:

Post-translační modifikace jsou zásadním mechanismem zvyšování komplexity proteomu. Umožňují vznik velkého množství forem jednotlivých proteinů, což je často potřebné pro regulaci jejich funkcí především v signálních kaskádách. Tím je možné dosáhnout přesných odpovědí nevyhnutelných pro regulaci složitých procesů. Wnt je vysoce konzervovaná a velice složitá signalizace, která je zásadní pro správný vývoj buněk a udržování homeostázy. Protein Dishevelled má úlohu klíčového regulátoru v téměř všech větvích Wnt signalizace. Tato práce se zaměřuje na identifikaci nové post-translační modifikace proteinu Dishevelled. Dokazuje, že enzymy podílející se na modifikaci interagují s tímto proteinem, což vede k jeho následné reverzibilní modifikaci. Kromě toho tato práce představuje předpokládané umístění popsané modifikace a pravděpodobný motiv potřebný pro úspěšnou modifikaci. Práce také poukazuje na možnou funkci nově objevené modifikace.

Abstract:

Post-translational modifications significantly increase proteome complexity allowing creation of different protein forms, which is fundamental for regulation of their broad functions. The modifications are often occurring in various signaling pathways to achieve fine-tuned responses necessary for regulation of complex processes. Wnt signaling is one of the highly conserved signaling pathways, required for various processes including cell development and maintenance of homeostasis. Dishevelled is a key regulatory protein which is involved in signaling events of almost all branches of the Wnt signaling pathway. This thesis deals with identification of a novel post-translational modification of Dishevelled. It shows that modifying as well as demodifying enzymes interact with Dishevelled resulting in its subsequent reversible modification. The thesis provides evidence about the localization of modification and suggests putative motif necessary for an effective modification. Furthermore, the thesis also provides insights into possible functions of the modification.
