Inducible cell systems in experimental oncology

Abstract:

Inducibilní systémy nám umožnují kontrolovat časovou i prostorovou expresi genů zájmu. Teoretická část bakalářské práce je věnována popisu vybraných inducibilních systémů a jejich modifikacím. Experimentální část práce se zabývá vlivem proteinu karboxypeptidázy B1 (CPB1) na migraci nádorových buněk MCF7 s využitím inducibilního systému. Zvýšená hladina proteinu CPB1 byla v dřívější studii zaznamenána u nádorů prsu subtypu luminální A metastazujícím do lymfatických uzlin oproti nemetastazujícím. K expresi proteinu CPB1 byl vybrán inducibilní Tet-On systém využívající doxycyklin jako induktor. Plazmidy kódující inducibilní Tet-On systém a gen CPB1 byly transfekovány do buněk MCF7 a kombinací antibiotické a monoklonální selekce byly kultivovány jednotlivé klony s nulovou bazální hladinou CPB1 a dostatečně vysokou indukovanou hladinou CPB1. U tří vybraných klonů a rodičovské linie MCF7 byl poté proveden test migrace metodou „scratch assay“ ve třech nezávislých opakováních. Po 24 h indukci doxycyklinem byly buňky rýhovány a byla stanovena hladina indukované CPB1 v čase 0 h. Monitorováním zarůstání rýhy po 24 a 48 h bylo zjištěno, že nejrychleji migrují rodičovské buňky MCF7 v porovnání s testovanými klony. Hladina CPB1 v průběhu experimentu klesala. Nebyl zaznamenán vliv exprese CPB1 ani doxycyklinu na rychlost migrace. V tomto experimentálním systému nemá zvýšená hladina proteinu CPB1 vliv na migraci buněk MCF7.

Abstract:

Inducible systems allow us to control the temporal and spatial expression of genes of interest. The theoretical part of the bachelor thesis is devoted to the description of selected inducible systems and their modifications. The experimental part of the thesis deals with the effect of CPB1 protein on the migration of MCF7 tumor cell line. Increased levels of CPB1 protein were previously observed in luminal A breast tumors subtype with metastases to lymph nodes compared to their non-metastatic counterparts. The inducible Tet-On system that utilizes doxycycline as an inducer was selected to express CPB1 protein. Plasmids encoding the inducible Tet-On system and the CPB1 gene were transfected into MCF7 cells, and single clones with no basal CPB1 level and sufficiently high induced CPB1 level were cultured by the combination of antibiotics and monoclonal selection. The three selected clones and the parental MCF7 line were then tested for migration by the scratch assay in three independent experiments. After 24 h period of induction with doxycycline, cells were scratched and the level of induced CPB1 was measured at time of 0 h. By monitoring the scratch after 24 and 48 h, the parental MCF7 cells were found to migrate more quickly compared to the tested clones. The level of CPB1 decreased during the experiment. No effect of CPB1 or doxycycline expression on migration rate was observed. In this experimental setting, CPB1 protein had no effect on the migration of MCF7 cells.
