Molecular regulation of early limb development

Abstract:

Vývoj končetin je souhrnem buněčných a molekulárních procesů vedoucích k vytvoření funkční identity končetin. Jejich přesná iniciace a koordinace je esenciálním faktorem při ustanovování a udržování aktivity hlavních signalizačních center končetin, která posléze řídí modelování finálního tvaru končetin. Každá i nepatrná porucha v této signalizaci může vést jednak k vytvoření oboustranné asymetrie končetin, ale i vzniku fatálních malformací končetin. Pro pochopení mechanizmu regulace vývoje končetin podél anterio-posteriorní osy je nejdříve nutná detailní identifikace klíčových molekul, které jsou zapojeny do jejich formování. Tato práce předkládá přehlednou celogenomovou expresní analýzu odhalující charakteristické rozložení řady genů podél anterio-posteriorní osy formujícího se základu končetin. Dále jsme prokázali rozdílný potenciál diferenciace anteriorních a posteriorních buněk do chondrocytové linie. Následně jsme se zaměřili na studium funkce vybraných ligandů WNT rodiny, která je nedílnou součástí vývoje končetin. Zatímco kanonická WNT signalizace vedla k alteraci chondrogeneze, nekanonické ligandy výrazně neovlivnily ranou chondrogenezi v in vitro podmínkách, avšak jejich aplikace do anteriorní či posteriorní části končetinových pupenů kuřecích embryí indukovala výrazné malformace křídel. Alterace WNT signalizace pomocí nové syntetické molekuly C59 naopak způsobila masivní indukci tvorby chrupavčité tkáně, jak v buněčných, tak v tkáňových kulturách. Ektopická aktivace FGF signalizace vedla k charakteristické inhibici chondrogeneze s alterovaným růstem chrupavčité složky vyvíjející se kosti v in vitro podmínkách. Aplikace FGF2 společně s nekanonickými WNT ligandy vedla k částečné obnově fenotypu, avšak jejich přímou interakci bude nutné ještě prokázat i na molekulární úrovni. Zapojení kanonické molekuly WNT3a a nekanonických WNT5a i WNT7a do procesu chondrogeneze a anterio-posteriorního formování končetin je dosud jen zčásti objasněno. Naše analýza jejich funkce během chondrogeneze a interakce s FGF rodinou povede k pochopení řady malformací končetin, které jsou charakteristické defekty podél A-P osy jako jsou polydaktylie, tak i defektů růstu dlouhých kostí do délky. Navíc nově identifikovaná funkce inhibitoru C59 by mohla být využita pro stimulaci chondrogeneze v tkáňovém inženýrství či pro regeneraci chrupavky.

Abstract:

Limb development consists of cellular and molecular processes, which are crucial for the establishment of limb identity. Determination and initiation of these processes is essential for the induction and maintenance of main signalling centres in the limb that later pattern final shape of limb structures. Disruption of these signals can lead to abnormal asymmetrical morphology of the limbs or fatal limb malformations. To understand the mechanisms contribution to the initiation of limb malformations along anterior-posterior axes, it will be first necessary to identify new molecular signals regulating A-P patterning. This master thesis determines the main molecular differences between anterior and posterior areas of the limb bud using microarray analyses. Moreover, the characteristic gene expression pattern was associated with differencesin cell cultures capacity to be able to differentiate into chondrocyte lines, which was observed in primary cell cultures established from the anterior and posterior mesenchymal cells. For functional test, we selected three members of WNT signalling family that are known to play a key role during limb development. While ectopic increase of canonical WNT signalling led to the alteration of chondrogenesis, non-canonical ligands did not significantly affected early chondrogenesis in in vitro conditions. Their application into the anterior or posterior areas of the limb buds in chicken embryos caused substantial malformations of limbs. A new inhibitor of WNT signalling C59 caused a massive production of cartilage tissue in cell as well as organ cultures. Ectopic activation of the FGF signalling triggered the inhibition of chondrogenesis and alteration of cartilage growth. The application of ectopic FGF ligand together with non-canonical WNT molecules resulted in a partial rescue of the phenotype but their interaction has to be necessary further analysed at the molecular level. The role of canonical WNT3a and non-canonical WNT5a or WNT7a in chondrogenesis and limb development along anterior-posterior axis has not been well determined yet. Our analysisfocused on the role of WNT signalling during chondrogenesis and its possible interaction with FGF signalling will help to understand the mechanisms contributing tospecific limb malformations along anterior-posterior axis including polydactyly or dwarfism phenotype. Moreover, a novel role of WNT inhibitor C59 could serve as a powerful tool to stimulate chondrocyte differentiation in tissue engineering or cartilage regeneration.
