Abstract:

Cílem teoretické části této práce bylo shrnout nejdůležitější organické sloučeniny síry vyskytující se v lidské plazmě, stručný popis jejich vzniku, metabolismu a jejich funkce ve fyziologických a patologických dějích probíhajících v lidském organismu. Dalším stěžejním tématem bylo prozkoumat, zda a případně jakým způsobem se v dnešní době hladina těchto látek stanovuje v klinické praxi. U klíčových sloučenin byl diskutován také potenciál implementace jejich stanovení do klinické praxe. V poslední části literární rešerše byly shrnuty elektrochemické metody použité v praktickém experimentu diplomové práce. Praktická část studie byla zaměřena na návrh a konstrukci elektrochemického senzoru vhodného pro simultánní stanovení redukované a oxidované formy glutathionu. Experiment zahrnoval výběr vhodného typu elektrody, její modifikaci, výběr elektrolytu a optimalizace jeho pH. Následně byla elektrochemicky stanovena koncentrace obou forem glutathionu, a to nejprve v elektrolytu a následně ve vzorku lidské plazmy. Paralelně s tímto experimentem probíhal ještě experiment s tištěnými elektrodami, které představují do praxe snáze převeditelnou platformu. Poslední částí praktického experimentu bylo zpracování dat a jejich interpretace.

Abstract:

The goal of the theoretical part of this thesis was to summarize the most important organic compounds of sulfur present in the human plasma, a brief description of their origin, metabolism, and their function in physiological and pathological processes ongoing in the human body. The following essential theme of the thesis was to analyze whether these compounds' level is nowadays being determined in the clinical practice and, if so, how. In the case of essential compounds, the potential of implementation of their determination in clinical practice was discussed. The electrochemical methods used in the practical part of this thesis were summarized in the last part of the literature review. The study's practical part focused on the proposal and construction of electrochemical sensors suitable for simultaneous determination of both reduced and oxidized glutathione forms. The experiment consisted of selecting a suitable type of electrode, its modification, the selection of electrolyte and optimization of its pH. Subsequently, the concentration of both forms of glutathione was determined, first in electrolytes and then in the human plasma sample. Simultaneously with this experiment, testing with screen printed electrodes (which represent an alternative, easier to put into practice platform) was performed. The last part of the practical experiment consisted of data processing and its interpretation.
