Molecular-genetic analyses of Carassius species in the central Europe

Abstract:

V oblasti střední Evropy se vyskytují dva druhy rodu Carassius: nativní karas obecný (Carassius carassius Linnaeus 1758) a nepůvodní karas stříbřitý (Carassius auratus Linnaeus 1758), který je považován za jeden z nejúspěšnějších invazivních druhů ve vodách Evropy. V oblasti biologie a taxonomie forem, které jsou souhrnně označovány jako „karas stříbřitý“, však stále existuje mnoho nejasností. V předkládané práci byla studována genetická diverzita komplexu karasa stříbřitého (Carassius auratus) technikami přímého sekvencování dvou úseků mitochondriální DNA: kontrolní oblasti a genu pro cytochrom b. Molekulárně-genetické analýzy prokázaly ve vodách České republiky a Slovenska přítomnost čtyř oddělených fylogenetických linií, které odpovídají čtyřem formám komplexu karasa stříbřitého. Jsou to formy Carassius auratus gibelio, Carassius auratus auratus, Carassius auratus langsdorfii a čtvrtá forma neznámé identity. Zjištěná vysoká genetická diverzita může mít souvislost s vysokou invazivní úspěšností karasa stříbřitého. Analýza čtyř mikrosatelitových markerů potvrdila hybridizaci mezi karasem obecným a formou Carassius auratus gibelio v oblasti dolního toku řeky Dyje. Analýza mitochondriální DNA prokázala, že jde o produkty jednosměrného křížení samců C. carassius a samic formy C. a. gibelio. Jedná se o první záchyt těchto hybridů na území České republiky. Analýza šesti mikrosatelitových markerů a sekvencování mitochondriální DNA populace karasa stříbřitého z umělého mokřadu v zátopovém pásmu řeky Moravy prokázaly koexistenci dvou klonálních linií karasa stříbřitého, odpovídajících formám Carassius auratus gibelio a Carassius auratus langsdorfii.

Abstract:

Two species of the genus Carassius occur in the central Europe: the native crucian carp (Carassius carassius Linnaeus 1758) and the non-indigenous silver crucian carp (Carassius auratus Linnaeus 1758), which is considered to be one of the most successful invasive species in the waters of the Europe. However, there is still much confusion in the field of biology and taxonomy of forms, which are collectively labeled as the „silver crucian carp“. In the proposed thesis, genetic diversity of the silver crucian carp (Carassius auratus complex) has been studied by methods of direct sequencing of two segments of the mitochondrial DNA: the control region and the cytochrome b gene. Molecular-genetic analyses have proven existence of four distinct phylogenetic lineages, which correspond to four forms of the silver crucian carp complex, in the waters of the Czech Republic and Slovakia. Those forms are Carassius auratus gibelio, Carassius auratus auratus, Carassius auratus langsdorfii and a fourth form of unknown identity. Uncovered high genetic diversity could relate to the high level of invasive success of the silver crucian carp. Analysis of four microsatellite markers has confirmed hybridization between the crucian carp and the Carassius auratus gibelio form in the lower reach of the Dyje River. Analysis of the mitochondrial DNA proved that those hybrids resulted from unidirectional hybridization between C. carassius males and C. a. gibelio females. Such hybrids have been found on the territory of the Czech Republic for the first time. Analysis of six microsatellite markers together with sequencing of the mitochondrial DNA in a silver crucian carp population from an artificial wetland in the Morava River floodplain have proven a coexistence of two clonal lineages of the silver crucian carp in the wetland. Those two lineages correspond to the Carassius auratus gibelio and Carassius auratus langsdorfii forms.
