Synthesis and cytotoxicity of caffeic acid derivatives

Abstract:

Kyselina kávová a některé její deriváty vykazují řadu biologických a farmakologických účinků. Odpovědnost za tyto účinky nese jejich specifická struktura. Cílem mé diplomové práce byla syntéza modelových sloučenin, konkrétně derivátů kyseliny kávové, které pak byly zkoumány jako nové ligandy aktivující obecné retinoidní receptory. Studované deriváty kávové kyseliny (methyl-, butyl-, decyl- a (E,E)-hexa-2,4-dienylester, (E)-3-(3,4-dimethoxyfenyl)propenová kyselina a její methylester) byly syntetizovány z kyseliny kávové a příslušného alkoholu. K metodě umožňující studium vlivů na retinoidní signálování byla použita buněčná linie P19/A15, transfekovaná genem exprimujícím luciferázu. Tato metoda umožňuje hodnocení toxicity látek narušující retinoidní signálování, k nimž patří antropogenní polutanty (ftaláty, polycyklické aromatické uhlovodíky, aj.), agrochemikálie nebo farmaceutika. Mezi výše zmíněnými studovanými deriváty kávové kyseliny nebyly nalezeny nové ligandy pro retinoidní receptory. Methylester kávové kyseliny, a to v obou geometrických izomerech, narušoval retinoidní signálování pouze v přítomnosti přirozeného ligandu (ATRA) pro retinoidní receptory. Potenciace ATRY na receptory byla pozorována i u (E)-azobenzenu, který byl zvolen jako výchozí systém pozorování chování izolovaných geometrických izomerů.

Abstract:

Caffeic acid and some of its derivatives display various biological and pharmacological effects. The specific of caffeic acid is responsible for the interactions with potential bioreceptors. The aim of my diploma thesis was synthesis of the different caffeic acid derivatives, which were investigated as potential new ligand for the retinoic acid receptors. A variety of caffeic acid derivatives (methyl-, butyl-, decyl- and (E,E)-hexa-2,4-dienyl ester, (E)-3-(3,4-dimethoxyphenyl)propenoic acid and the corresponding methyl (E)-3-(3,4-dimethoxyphenyl)propenoate) was synthesized from caffeic acid and an alcohol. A cell line P19/A15, transfected with luciferase gene, was used as material for testing the effect of prepared derivatives on retinoic signall pathway. This method is generally used for examination of compounds such as anthropogenic pollutant (phthalates, polycyclic aromatic hydrocarbons, etc.), agrochemicals or pharmaceutics, which affect a retinoid signalling. None of the investigated derivatives was found to be a new ligand for retinoic acid receptores. Both geometric isomers (E and Z) of methyl ester caffeic acid affects retinoic signall patway only in presence of nature ligand (ATRA) for retinoic receptor. Potentiation of ATRY for the receptors was also observed in presence (E)-azobenzene, which was then chosen as a primary system for comparing the separated geometric isomers (E and Z).
