Testing Of Cellular Nanotoxicity

Abstract:

Diplomová práce se zabývá testováním cytotoxicity nanomateriálů. Teoretická část se věnuje suspenzním buněčným liniím, dále je zde popsána toxicita nanomateriálů, a v závěru jsou uvedeny testy, které se používají k hodnocení cytotoxicity nanomateriálů. Experimentální část je zaměřena na testování cytotoxicity vybraných nanomateriálů pomocí bioanalytických a mikroskopických metod na suspenzní buněčné linii Jurkat. Pro experimenty jsme zvolili nanočástice TiO2 P25, TiO2 rutil a uhlíkové nanotrubičky SWCNTs a MWCNTs. Potencionální toxicitu jsme hodnotili po 24 a 48 hodinách inkubace buněk Jurkat s nanomateriály o koncentraci 0-500 ?g/mL. Vliv vybraných nanomateriálů na životaschopnost buněk jsme hodnotili pomocí WST-1 testu a vliv na intracelulární koncentraci glutathionu jsme detekovali pomocí fluorescenční sondy monochlorobiman. Morfologii buněk inkubovaných s nanočásticemi nebo nanotrubičkami jsme pozorovali pod invertovaným mikroskopem ve fázovém kontrastu. V závěru práce jsou prezentovány výsledky a je zhodnocena toxicita nanomateriálů.

Abstract:

The aim of this diploma thesis is testing of cellular nanotoxicity. First, the theoretical part of the thesis deals with the suspension cell lines, then the toxicity of nanomaterials is described. Finally, the assays used to evaluate the cytotoxicity of nanomaterials are described. The experimental part is focused on testing of the cytotoxicity of some nanomaterials using bioanalytical and microscopic methods in the Jurkat cell line. For experiments we chose TiO2 P25 and rutil nanoparticles together with carbon SWCNTs and MWCNTs nanotubes. The potential cytotoxicity was assessed in Jurkat cells after 24 and 48 hours of incubation with nanomaterials at concentrations 0-500 ?g/mL. The effect of presented nanomaterials on cell viability was evaluated using the WST-1 test and the effect on the intracellular level of glutathione was detected using the optimized monochlorobimane assay. We observed the morphology of Jurkat cells incubated with nanoparticles or nanotubes using an phase contrast inverted microscope. At the end of the thesis, the results are presented, and toxicity of nanomaterials is evaluated.
