Optimalization of chloroplasts isolation technique for lichen photobionts of the genus <I>Tebouxia</I>

Abstract:

Náplní této diplomové práce je najít nejvhodnější metodu izolace chloroplastů z lišejníkových fotobiontů. Izolace intaktních chloroplastů je nezbytná pro další studium fotosyntetických procesů. Tato diplomová práce obsahuje základní informace o lišejnících. Další velká část teoretické části práce je věnována lišejníkovým fotobiontům. Jako modelový organismus byla zvolena zelená řasa rodu Trebouxia, která je také popsána v teoretické části práce. Tato teoretická část může posloužit jako výukový nebo podpůrný materiál pro žáky středních škol. Větší důraz je kladen na praktickou část této práce, kde jsme modifikovali již publikované postupy izolace chloroplastů tak, aby byly metody vhodné k izolaci chloroplastů pro tento typ experimentálního materiálu. Aktivitu vyizolovaných chloroplastů jsme zkoumali měřením parametrů fluorescence chlorofylu FV/FM, FM, F0, parametry rychlé indukční kinetiky VJ, Vi a Area. Celá práce je doplněna fotodokumentací průběhu izolačních metod. V rámci této diplomové práce byla v laboratoři fyziologie fotosyntézy, oddělení fyziologie a anatomie rostlin zavedena metoda izolace chloroplastů ze suspenzí kultur zelených řas.

Abstract:

The content of this diploma work is to find out the most suitable method of isolation of chloroplasts from lichen photobionts. The isolation of intact chloroplasts is necessary for next study of photosynthetic process. This diploma work contains basic information about of lichens. The next large part of the theoretic part of this diploma work is dedicated to a lichen photobionts. As a model organism was chosen green algae from the genus Trebouxia that is described in the theoretical part of the diploma work. This theoretical part may help as an instructional or supporting material for students of secondary schools. The bigger emphasis is laid out for practical part of this diploma work, where we modified already published procedures of isolation of chloroplasts in the way to be methods suitable to isolation of chloroplasts for this kind of experimental material. The activity of isolated chloroplasts we checked before measuring parameters of fluorescence chlorophyll FV/FM, FM, F0, parameters of fast inductive kinetics VJ, Vi and Area. The whole diploma work is fulfilled with documentation of progress isolation methods. In the context of this diploma work was in the laboratory of photosynthesis, department of plant physiology and anatomy implemented method of isolation of the suspension cultures of green algae.
