microRNAs as indicators of human diseases

Abstract:

MicroRNA molekuly jsou důležitými regulátory genové exprese, a to jak na posttranskripční, tak na transkripční úrovni. Od jejich relativně nedávného objevu bylo zjištěno, že se podílejí na velkém množství buněčných procesů, čímž silně ovlivňují vývoj i život jedince. Jejich deregulovaná exprese navíc souvisí se vznikem a průběhem mnohých lidských onemocnění. Tyto malé nekódující molekuly RNA taktéž disponují schopností přesouvat se do jakéhokoli místa na těle, pomocí mezibuněčných spojů a oběhové soustavy. Proto jsou detekovatelné jak z lidských tkáních, tak z tělních tekutin. Díky těmto skutečnostem se microRNA molekuly staly středem zájmu mnohých studií, které zkoumají jejich potenciál stát se neinvazivními diagnostickými markery pro včasnou detekci různých onemocnění. V současné době se uvažuje o molekulách microRNA taktéž jako o nových potenciálních terapeutických cílech, pomocí kterých by se v blízké budoucnosti dala různá onemocnění léčit s mírnějšími vedlejšími účinky, či by se pomocí nich napomohlo regeneraci tkání po vážných úrazech. Tato bakalářská práce je proto zaměřena na microRNA molekuly, jenž s lidskými chorobami souvisí a je studován jejich diagnostický potenciál pro metabolické onemocnění diabetes mellitus 2.typu, nádorová onemocnění (konkrétně pro rakovinu prsu a rakovinu plic), atopickou dermatitidu a Alzheimerovu chorobu.

Abstract:

MicroRNAs are important regulators of gene expression, both at the post-transcriptional and transcriptional levels. Since their relatively recent discovery, they have been found to be involved in a large number of cellular processes, thus strongly influencing the development and life of the individual. Moreover, their deregulated expression has been linked to the onset and progression of many human diseases. These small non-coding RNA molecules also have the ability to transport to any location on the body, using intercellular junctions and the circulatory system. Therefore, they are detectable from both human tissues and body fluids. Due to these facts, microRNAs have become the centre of interest of many studies investigating their potential to become non-invasive diagnostic markers for early detection of various diseases. Currently, microRNA molecules are also being considered as new potential therapeutic targets that could be used in the near future to treat various diseases with milder side effects or to aid tissue regeneration after severe injuries. Therefore, this thesis focuses on microRNAs related to human diseases and their diagnostic potential for metabolic disease type 2 diabetes mellitus, cancer (specifically breast and lung cancer), atopic dermatitis and Alzheimer's disease is being studied.
