Characterization of the variable part of coagulase-negative staphylococci genome

Abstract:

Koaguláza-negativní stafylokoky (KNS) jsou grampozitivní koky známé především jako komenzálové žijící na kůži a sliznicích člověka a zvířat. Velmi často jsou ale tyto bakterie také oportunními patogeny a způsobují širokou škálu onemocnění zahrnujících převážně nozomiální infekce zavedených cizích těles, ale také infekce krevního řečiště, kloubů a močového traktu u zdravých jedinců. Problematická je u nich léčba kvůli jejich velmi časté rezistenci k antimikrobiálním látkám, která se šíří také na jiné patogenní bakterie prostřednictvím mobilních genetických elementů (MGE). Přes velký význam KNS jako zdrojů rezistence k antibiotikům je tato oblast stále nedostatečně prozkoumána. Disertační práce navazuje na dosud známé poznatky o MGE u stafylokoků a rozšiřuje je. V práci jsou charakterizovány plazmidy, bakteriofágy a profágy, chromozomové ostrovy a další MGE a geny na nich nesené a je detailně zkoumán jejich přenos. Práce charakterizuje velkou variabilitu plazmidů KNS, jejich příbuznost s jinými KNS plazmidy i s plazmidy Staphylococcus aureus a potvrzuje možnost přenosu těchto elementů na jiné druhy. Práce dále přináší detailní charakteristiku bakteriofágů Staphylococcus epidermidis a průkaz jejich role v přenosu plazmidů a chromozomových ostrovů, které mohou být nositeli genů rezistence. Popsáno je také široké spektrum MGE u nedávno objeveného koaguláza-negativního druhu Staphylococcus petrasii s poukazem na jejich roli v šíření genů rezistence k antibiotikům a genů virulence mezi stafylokoky. Výsledky výzkumu byly zahrnuty do tří experimentálních publikací a disertační práce je zpracována ve formě komentovaného souboru těchto publikací.

Abstract:

Coagulase-negative staphylococci (CoNS) are gram-positive cocci mostly known as commensals on the skin and mucous membranes of humans and animals. These bacteria can, however, very often act as opportunistic pathogens and cause a wide range of diseases, including mainly nosomial infections of established foreign bodies, but also infections of the bloodstream, joints and urinary tract in healthy individuals. Treatment of these diseases is problematic due to their very frequent resistance to antimicrobial agents, which is also spread to other pathogenic bacteria via mobile genetic elements (MGEs). Despite the significance of CoNS as a source of antibiotic resistance, this area is still not well explored. The dissertation builds on the previous findings about MGEs in staphylococci and expands it. Plasmids, bacteriophages and prophages, chromosomal islands and other MGEs, as well as genes carried on them, are characterized and their transmission is investigated in detail. The work characterizes the great variability of CoNS plasmids, their relatedness to other CoNS plasmids and plasmids of Staphylococcus aureus, and confirms the possibility of transfer of these elements to other species. The work also provides a detailed description of bacteriophages in Staphylococcus epidermidis and evidence of their role in the transfer of plasmids and chromosomal islands, which may be carriers of resistance genes. A broad spectrum of MGEs in the recently discovered coagulase-negative species Staphylococcus petrasii is also described, with reference to their role in the spread of antibiotic resistance and virulence genes among staphylococci. The results of this research were included in three research articles and the dissertation is presented in the form of a commented collection of these publications.
