Measurement of cholinesterase inhibitors using smartphone

Abstract:

Cholinesterases are a group of enzymes that play a key role in hydrolysis of the neurotransmitter acetylcholine. They also function as target for the effects of carbamate and organophosphate insecticides, which affect the natural behaviour of cholinesterases, because they cause their inhibition. The purpose of this bachelor thesis was measuring the enzyme activity of acetylcholinesterases and the effect of the used inhibitor on its physiological function using chromogenic and fluorogenic substrates. For this purpose, the analytical determination used a smartphone camera as an optical element suitable for tracking color changes. Measuring the activity of enzymes was also verified with the use of laboratory instrumentation, fluorescent scanner and a standard method based on a spectrophotometer. Enzyme was fixed on a chosen, suitable indicator strip. The color changes of the uninhibited and inhibited samples were compared and evaluated with the use of a freely accessible programme ImageJ for processing of the taken pictures. In the experimental part of this thesis, it was discovered, that the fluorescent scanner and the smartphone camera are usable for analytical determination of enzyme activity.

Abstract:

Cholinesterasy jsou skupinou enzymů, které sehrávají klíčovou roli při hydrolýze neurotransmiteru acetylcholinu. Představují také cílové místo účinku karbamátových a organofosfátových insekticidů, které zasahují do přirozeného fungování cholinesteras, protože způsobují jejich inhibici. Cílem této bakalářské práce bylo měření enzymové aktivity acetylcholinesterasy a vlivu použitého inhibitoru na její fyziologickou funkci pomocí chromogenních a fluorogenních substrátů. V analytickém stanovování se za tímto účelem ověří použitelnost fotoaparátu smartphonu jako optického prvku vhodného ke sledování barevných změn. Měření aktivity enzymů bylo ověřené i pomocí laboratorní instrumentace, fluorescenčním skenerem a standardně využívanou metodou pomocí spektrofotometru. Stanovování proběhlo na zvoleném vhodném indikačním proužku, na kterém se barevné změny neinhibovaných a inhibovaných vzorků navzájem porovnávaly a vyhodnocovaly pomocí volně dostupného programu ImageJ pro zpracování obrázků. V experimentální části práce bylo zjištěno, že fluorescenční skener a fotoaparát smartphonu jsou využitelné při analytickém stanovování enzymatické aktivity.
