Effect of synonymous mutations on pre-mRNA splicing of the LDLR exon 4

Abstract:

Cleavage of non-coding parts - introns from pre-mRNA represents the basic step of gene expression, which is called pre-mRNA splicing. This process depends on numerous additional splice site regulatory elements and on a network of interactions between them, which increases the opportunity for mutations and improper regulation that causes diseases. In this thesis I deal with mutations in the LDLR gene, which cause the most common monogenic disease in population ̶ familial hypercholesterolemia. To verify the effect of three synonymous mutations on pre-mRNA splicing, we used minigen constructs containing exon 4 of the LDLR gene that were expressed in human tumoral lines in combination with in silico predictors. The results of the minigene analysis showed a non-significant amount of aberrant splice forms in all variants, but only c.685C>T a c.694C>T showed slightly increased exon skipping in comparison with the standard sequence. However, the significance of synonymous mutations should not be underestimated, as they are often the cause of severe diseases.

Abstract:

Vyštiepenie nekódujúcich častí - intrónov z pre-mRNA predstavuje základný krok génovej expresie, ktorý sa nazýva zostrih pre-mRNA. Tento proces závisí na početných prídatných regulačných elementoch zostrihu a na zložitej sieti interakcií medzi nimi, čo zvyšuje príležitosť pre mutácie a nesprávnu reguláciu, ktorá spôsobuje ochorenie. V tejto práci sa zaoberám mutáciami v géne LDLR, ktoré sú príčinou najčastejšieho monogénneho ochorenia v populácii ̶ familiárna hypercholesterolémia. Pre overenie efektu troch synonymných mutácii na zostrih pre-mRNA boli použité minigénové konštrukty obsahujúce exón 4 génu LDLR, ktoré boli exprimované v ľudských nádorových líniách, v kombinácií s in silico predikčnými nástrojmi. Výsledky minigénovej analýzy ukázali vznik nesignifikantného množstva aberantných zostrihových foriem u všetkých variant, no len u c.685C>T a c.694C>T došlo k mierne zvýšenému exon skippingu oproti štandardnej sekvencii. Napriek tomu by problematike týkajúcej sa synonymných mutácií mala byť venovaná dostatočná pozornosť, keďže existujú dôkazy, že sú dôvodom vzniku vážnych ochorení.
