Long non coding RNA molecules

Abstract:

Dlouhé nekódující molekuly RNA (lncRNA) tvoří skupinu nekódujících molekul s délkou větší než 200 nukleotidů. Tyto lncRNA, jenž jsou lokalizovány v jádře a cytosolu, jsou transkribovány pomocí RNA polymerázy II. Nicméně chybějící otevřený čtecí rámec jim neumožňuje translaci. Podrobují se různým transkripčním úpravám, například pre-RNA sestřihu a polyadenylaci. Mnohé vykazují epigenetické vlastnosti podobně jako geny kódující proteiny, například trimetylaci histonů H3K4 na 5´konci a trimetylaci histonů H3K36 uprostřed genu. Evoluční zachování lncRNA je možné shrnout do čtyř kategorií: sekvence, struktura, funkce a transkripce synténních genů. Dále jsou v práci popsány čtyři archetypy („signals“, „decoys“, „guides“, „scaffolds“), pomocí kterých lze určit molekulární funkce lncRNA. K objasnění původu a funkce též přispívá kapitola zabývající se rozdělením lncRNA z různých aspektů. Závěrečná část práce přibližuje roli lncRNA jako markeru pro detekci onemocnění. Konkrétně je zde popsán proces molekulárního výzkumu lncRNA PCA3 jako biomarkeru karcinomu prostaty s využitím v klinické praxi.

Abstract:

Long non-coding RNA molecules (lncRNA) are a group of non-coding RNA (ncRNA) longer than 200 nucleotides. These lncRNA are localized in the nucleus and cytosol. They are transcribed by RNA polymerase II. However, the lack of an open reading frame does not enable them to undergo translation. They undergo different transcription modifications, e.g. pre-RNA splicing and polyadenylation. Many of them display epigenetic features common to protein-coding genes, such as trimethylation of histone H3K4 at the 5´end and trimethylation of histone H3K36 in the middle of gene. Evolutionary conservation of lncRNA can be summarized in four cathegories: sequence, structure, function and syntenic transcription. Furthermore, this thesis describes four archetypes (signals, decoys, guides, scaffolds), which can be used to identify molecular function of lncRNA. To clarify the origin and function, a chapter dealing with the division of lncRNA from different perspectives was included. The final part of this thesis mentions the role of lncRNA as a marker for detection of diseases. Specifically, a process of molecular research of lncRNA PCA3 as a biomarker for prostate cancer with using in clinical practice is described.
