Verification of the methodology for quantitative determination of chlorophylls and carotenoids using high performance liquid chromatography.

Abstract:

Znalost obsahu a zastoupení jednotlivých fotosyntetických pigmentů v rostlinách představuje cennou informaci, od které můžeme odvodit řadu dalších charakteristik týkajících se fyziologického stavu rostliny nebo např. úrovně stresu, kterému je rostlina vystavena. Cílem této práce bylo provedení kalibrace a validace dvou zavedených metod pro stanovení obsahu fotosyntetických pigmentů pomocí vysokoúčinné kapalinové chromatografie s optickým absorpčním detektorem typu diodového pole. Pro práci byly zakoupeny ve formě standardů dva základní fotosyntetické pigmenty, které se vyskytují ve vyšších rostlinách i dalších organismech (chlorofyl a, chlorofyl b). Pro tyto pigmenty byly stanoveny validační parametry přenositelnost, správnost, přesnost, selektivita, mez detekce a kvantifikace, linearita a byly vytvořeny kalibrační křivky. Následně byly nově zavedené kalibrační křivky využity ke stanovení obsahu chlorofylů v extraktech z rostlin Arabidopsis thaliana k porovnání výsledků z obou metod. Výsledky z obou zavedených metod ukázaly, že tyto metody nezpůsobují přenos do dalších měření, jsou správné, přesné a selektivní (z hlediska čistoty stanovovaných látek dle použité metody vyhodnocení). Při stanovení nových kalibračních křivek byly naměřené výsledky hodnoceny jako lineární. Dále byly také testovány dva typy vyhodnocení kalibračních křivek, kdy výsledky ukázaly jen velmi malé rozdíly při stanovení správnosti obou těchto typů vyhodnocení (stanovení z průměru naměřených hodnot, stanovení ze všech naměřených hodnot). K příslušným kalibračním křivkám byly vypočítány meze detekce a meze kvantifikace. Při otestování aplikovatelnosti byly použity kalibrační rovnice stanovené z průměru naměřených hodnot, jelikož lépe splňovaly parametr linearity než hodnoty stanovené pomocí všech naměřených hodnot. Stanovení obsahu pigmentů pomocí kalibračních křivek (HPLC vyhodnocení) byly v rámci aplikovatelnosti porovnány s výsledky spektrofotometrického stanovení a s výsledky stanovenými pomocí konverzních faktorů (taktéž HPLC vyhodnocení). Porovnáním těchto metod vyhodnocení bylo zjištěno, že hodnoty získané konverzními faktory se podstatně liší od hodnot získaných kalibrační křivkou či pomocí absorpčního spektrofotometru. Celkově jsou nově zavedené kalibrační křivky v případě obou metod vhodné ke stanovení chlorofylů, a v případě menšího ředění (dostatečné koncentrace) i ke stanovení vybraných derivátů chlorofylů.

Abstract:

Knowledge of the content of individual photosynthetic pigments in plants is valuable information from which we can derive a number of other characteristics related to the physiological state of the plant or, for example, the level of stress to which the plant is exposed. The aim of this work was to perform a calibration and validation of two established methods for determining the content of photosynthetic pigments using high-performance liquid chromatography. Two basic photosynthetic pigments found in higher plants and other organisms (chlorophyll a, chlorophyll b) were purchased as standards for the work. For these pigments, the validation parameters transmission, accuracy, precision, selectivity, limit of detection and quantification, linearity were determined and calibration curves were created. Subsequently, the newly introduced calibration curves were used to determine the chlorophyll content in extracts from Arabidopsis thaliana plants to compare the results from both methods.The results from both methods showed that they do not cause transfer to other measurements, are correct, accurate and selective (in terms of the purity of the determined substances according to the evaluation method used). When determining the new calibration curves, the measured results were evaluated as linear. Furthermore, two types of evaluation of calibration curves were also tested, where the results showed only very small differences in determining the accuracy of both of these types of evaluation (determination from the average of measured values, determination from all measured values). Limits of detection and limits of quantification were calculated for the respective calibration curves. When testing the applicability, the calibration equations determined from the average of the measured values were used, as they better met the linearity parameter than the values determined using all the measured values. The determination of pigment content using calibration curves (HPLC evaluation) were compared within the scope of applicability with the results of spectrophotometric determination and with the results determined using conversion factors (also HPLC evaluation). By comparing these methods, it was found that the values obtained by conversion factors differ significantly from the values obtained by the calibration curve or by means of an absorption spectrophotometer. Overall, the newly introduced calibration curves are suitable for the determination of chlorophylls, and in the case of a sufficient concentration also for the determination of selected chlorophyll derivatives.
