Is there any effect of the presence of father on the House mouse offspring exploratory behaviour?

Abstract:

Dospívání je u savců kritickým obdobím pro vývoj chování. Studium vlivu prostředí, v němž se jedinec po tuto dobu nachází, je proto důležitou součástí výzkumu ontogeneze různých fyziologických, psychologických a behaviorálních aspektů. V sociálních skupinách myši domácí funguje přísná hierarchie, jejíž důsledkem je také tlak, kterým působí dominantní jedinci na ostatní členy dému. Tento tlak se pokládá za silný motivátor k opuštění skupiny a k následné disperzi. Proto jsme se pokusili objasnit, zda má přítomnost samce-otce v době narození a dospívání efekt na explorační chování potomků. Jelikož se dva nejrozšířenější poddruhy myši domácí v míře agresivity a strategii explorace liší, zajímalo nás, jestli bude případný efekt u obou taxonů stejný. Obecně jsme také chtěli objasnit, nakolik si v tomto směru může dovolit experimentátor tento faktor (odchov mláďat izolovanou samicí vs. “v rodine”) v designu pokusu ignorovat. Celkem bylo testováno 21 samců z první generace potomků divokých jedinců, narozených v zajetí (12 jedinců Mus musculus musculus, 11 jedinců M. m. domesticus) a 40 samců druhé generace (20 jedinců M. m. domesticus, 20 M. m. musculus) ve dvou experimentálních režimech – s otcem a bez otce. Explorační chování jsme testovali ve standardním Open-field testu. Vliv přítomnosti/absence otce na explorační profil potomků se nám nepodařilo prokázat. Explorační chování testovaných jedinců navíc neodpovídá poddruhově specifickým projevům, jež jsme pozorovali v předešlých studiích. Zdá se, že ve věku 55 – 56 dnů, což bylo stáří námi testovaných zvířat, ještě není behaviorální profil ustálený a případný efekt otce se zatím nemusí projevit. Věk testovaných zvířat se tedy jeví jako závažný faktor, jeho pečlivé plánování a přesná specifikace by neměly být v metodice experimentů zanedbávány.

Abstract:

Adolescence is a critical period for the development of behavior in mammal species. Studying the influence of the environment, in which the subadult individual develops, is thus an important for understanding the ontogeny of many physiological, psychological and behavioral aspects. In the house mouse social group, a rigorous hierarchy is usually established. Dominant individuals exerted agonistic behaviour and social pressure to other members of the deme (social groups). It is assumed, that this pressure is an important motivator, especially for the young animals, to leave the group and to start the dispers. In this study we investigated the presence of the male at time of birth and adolescence as a factor possibly modulating exploratory behavior of the offspring. As the two most common house mouse subspecies differ in their aggression and exploration patterns, we were curious whether the effect, if present, is identical for both taxa. In more general context, we wanted to estimate to what extent can the experimenter afford to ignore this factor (rearing animals with isolated females vs. " whole familly"). In total, 21 males of the first generation offspring of wild individuals were tested (12 individuals of Mus musculus musculus, 11 of M. m. domesticus) and 40 males of the second generation (20 M. m. domesticus, 20 M. m. musculus). The second generation males were reared in two experimental modes - with and without the presence of father. We tested the exploratory behavior in a standard open-field test. We did not detect any effect of the presence/absence of the father on exploratory profile of offspring. Unfortunatelly, it seems that the behavioral profile is not yet stable in the age of 55 - 56 days (age of testing). Moreover, we were also unable to detect consistent sub-specific differences, previously reported in other studies. This idicates, that the age of tested animals is important factor, and careful planning and thorough description should not be neglected in the methodology of experiments.
