Detection of extracellular traps in human leukocytes

Abstract:

V posledních letech se do popředí zájmu stále více dostává proces aktivní buněčné smrti neutrofilů zvaný NETóza (z angl. NETosis). Při NETóze neutrofil ejektuje do extracelulárního prostoru rozvolněná chromatinová vlákna asociovaná s granulárními proteiny s cílem usmrtit nebo imobilizovat infekční agens v místě zánětu. První informace o NETóze pocházejí z roku 2004 a od té doby je tento proces aktivně studován. Bohužel však ve většině studií je NETóza detekována pomocí fluorescenční mikroskopie, což je metoda, ke které mají přístup jen některé laboratoře. Z tohoto důvodu je snaha o nalezení dalších analytických přístupů k detekci NETózy. V poslední době vzrůstá zájem o detekci NETózy především pomocí flow cytometrie. Tato diplomová práce se zabývá detekcí NETózy u neutrofilů získaných z různých materiálů – neutrofily z plné krve (PK), izolované granulocyty a neutrofily o nízké hustotě (LDN). NETóza je detekována pomocí dvou přístrojových metod – flow cytometrie a měření intenzity fluorescence multimodální čtečkou mikrotitračních destiček EnSight od firmy PerkinElmer. Teoretická část této diplomové práce se zabývá úlohou neutrofilů v imunitním systému a podrobným popisem procesu NETózy včetně negativních dopadů na organismus při deregulaci tohoto procesu. Praktická část se zabývá detekcí NETózy pomocí dvou výše zmíněných metod. V praktické části je kladen důraz na nalezení optimální kombinace měřených NETotických markerů na povrchu NETotických neutrofilů. Dále se praktická část zabývá vlivem NETózu-indukujících stimulancií (ionomycin a PMA) na míru exprese (MFI) NETotických znaků. Dále je zjišťováno, zda existuje rozdíl v expresi NETotických znaků, a také zda existují rozdíly v procentuálním zastoupení NETózujících neutrofilů mezi neutrofily získanými z různých materiálů (LDN, PK, IG). V poslední části jsou porovnávány výsledky měření NETózy pomocí flow cytometrie a měření intenzity fluorescence multimodální čtečkou mikrotitračních destiček EnSight. Hlavním zjištěním je, že NETotické znaky Sytox Green (SG), Live Dead (LD), myeloperoxidáza (MPO) a citrulinovaný histon H3 (HisH3) jsou vhodnými znaky k detekci NETózy pomocí flow cytometrie.

Abstract:

In recent years, the process of active cell death of neutrophils known as NETosis, has been gaining increasing attention. During NETosis, neutrophils eject decondensed chromatin fibers along with granular proteins into the extracellular space, aiming to kill or immobilize infectious agents at the site of inflammation. The first information about NETosis dates back to 2004, and since then, this process has been actively studied. However, in most studies, NETosis is detected using fluorescence microscopy, a method accessible to only a few laboratories. Due to this reason, there is an effort to find alternative analytical approaches for NETosis detection. Lately, there is a growing interest in detecting NETosis, primarily using flow cytometry. This master's thesis focuses on detecting NETosis in neutrophils obtained from various sources – neutrophils from whole blood (WB), isolated granulocytes (IG) and Low Density Neutrophils (LDN). NETosis is detected using two instrumental methods – flow cytometry and measurement of fluorescence intensity with the EnSight multimode plate reader from PerkinElmer. The theoretical part of this thesis discusses the role of neutrophils in the immune system and provides a detailed description of the NETosis process, including its negative effects on the organism when this process becomes dysregulated. The practical part deals with the detection of NETosis using the two methods mentioned above. The practical section emphasizes finding the optimal combination of measured NETotic markers on the surface of NETotic neutrophils. Additionally, the practical part examines the impact of NETosis-inducing stimulants (ionomycin and PMA) on the expression level (MFI) of NETotic markers. The practical part also investigates whether there are differences in the expression of NETotic markers and whether there are variations in the percentage of NETosing neutrophils among neutrophils obtained from different sources (LDN, WB, IG). In the final section, the results of NETosis measure-ments using flow cytometry are compared with measurements of fluorescence intensity using the EnSight multimode plate reader. The main finding is that NETotic markers, such as Sytox Green (SG), Live Dead (LD), myeloperoxidase (MPO), and citrullinated histone H3 (HisH3), are suitable for NETosis detection using flow cytometry.
